Hemodynamic changes in a rat parietal cortex after endothelin-1-induced middle cerebral artery occlusion monitored by optical coherence tomography

Abstract: A blockage of the middle cerebral artery (MCA) on the cortical branch will seriously affect the blood supply of the cerebral cortex. Real-time monitoring of MCA hemodynamic parameters is critical for therapy and rehabilitation. Optical coherence tomography (OCT) is a powerful imaging modality that can produce not only structural images but also functional information on the tissue. We use OCT to detect hemodynamic changes after MCA branch occlusion. We injected a selected dose of endothelin-1 (ET-1) at a depth… Show more

“…In this study, a spectral-domain OCT (SD-OCT) system was employed, which is similar to our previous work. 11,12 Briefly, a superluminescent diode with a central wavelength of 1310 nm and a bandwidth of 65 nm was used as a light source, providing an ∼8.9-μm axial resolution in the tissue. In the sample arm, an achromatic lens with 30-mm focal length was used to achieve theoretical lateral resolution of ∼9 μm.…”

“…The model of the focal cerebral ischemia used in our experiments was based on the previously described endothelin-1- induced middle cerebral artery occlusion (MCAO) model. 12 The stereotaxic coordinates of the ET-1 injection were −0.83-mm anterior, −3.36-mm lateral, and 1.0-mm ventral relative to bregma. A dose of 3-μL ET-1 (dilute to 100 pmol/L in 0.9% normal saline) was injected at 1 μL∕ min.…”

“…This OCT system uses near‐infrared light. Because the frequencies and bandwidths of infrared light are orders of magnitude higher than medical ultrasound signals, which results in greater image resolution, OCT enables imaging capillaries in mice .…”

“…OCT angiography (OCTA) is a functional extension of OCT and is able to provide 3D blood perfusion map in tissue down to capillary level without a need for contrast agents . OCTA has been successfully used to image micro‐vascular networks in a variety of tissues in vivo, for example, brain , retina , skin , skeletal muscles and microvascular anastomosis .…”

“…The model of cerebral ischemia used in our experiments was based on endothelin-1-induced MCAO model described previously [31]. The stereotaxic coordinates of the ET-1 (Shanghai Jianglai industrial Limited by Share Ltd. China) injection were 0.9 mm anterior, 5.2 mm lateral and 8.7 mm ventral relative to bregma.…”
