Hemicyanine‐Based Near‐Infrared Activatable Probes for Imaging and Diagnosis of Diseases

Abstract: Molecular activatable probes with near‐infrared (NIR) fluorescence play a critical role in in vivo imaging of biomarkers for drug screening and disease diagnosis. With structural diversity and high fluorescence quantum yields, hemicyanine dyes have emerged as a versatile scaffold for the construction of activatable optical probes. This Review presents a survey of hemicyanine‐based NIR activatable probes (HNAPs) for in vivo imaging and early diagnosis of diseases. The molecular design principles of HNAPs toward… Show more

“…1a). P-CyPt comprises (1) a NIR merocyanine fluorophore (mCy) 26,27 capped with an ALP-recognition phosphate group (PO 3 H) 28 , (2) a GSH-reducible CDDP prodrug (Pt(IV)) 29,30 , and (3) a hydrophobic D -Phe-D -Phe (FF) dipeptide 31 , which shows low NIR FL, low PA signal and low cytotoxicity. After systemic administration, P-CyPt as a small molecule can extravasate and penetrate deeply into tumor tissues (Fig.…”

“…While nanoparticle formulations can potentially offset these drawbacks, it is preferable to develop complementary SWIR dyes that do not require encapsulation for in vivo imaging. In this study, we strategically remodeled the hemicyanine dye (HD) 14,15 (a common NIR platform) for multimodal SWIR FL and PA imaging. Of note, complementary remodeling efforts have yielded PA and NIR-II analogs.…”
