Heme−Peptide Models for Hemoproteins. 1. Solution Chemistry of N-Acetylmicroperoxidase-8

Abstract: An improved method for the preparation of the heme octapeptide acetyl-MP8, obtained by proteolysis of horse heart cytochrome c, is described. AcMP8 obeys Beer's law at pH 7.0 in aqueous solution up to a concentration of 3 x 10(-)(5) M. The self-association constant measured at 25 degrees C (log K(D) = 4.04) is an order of magnitude lower than that for MP8, reflecting the role of the N-acetyl protecting group in abolishing intermolecular coordination. However, AcMP8 does form pi-stacked dimers in aqueous soluti… Show more

“…The b-type waterhemes have an Em,sol of -120 mV, while the a-type heme in cytochrome c oxidase has an Em,sol of -20 mV. Hydroxyl-hemes have Em,sols 80 mV lower than the water-hemes (72,73), consistent with the 1.3 pKa unit shift between the pKas of ferric and ferrous aquo-heme (70,71). b The pKa and Em of the aquo-heme a3 is calculated with CuA oxidized and heme a and CuB reduced to mimic the experimental pKa measurements in the CO photolyzed mixed-valence complex (53).…”

“…To couple proton uptake to BNC reduction, the hydroxyl pK a must be lower than the solution pH in the fully oxidized state and higher following reduction. The pK a,sol for deprotonation of an oxidized Hisaquo-heme has been measured to be 9.6 (86), while that of an oxidized Cu(II) with 3 His ligands is estimated to be 9.4 (88,89). In the fully oxidized protein, MCCE Monte Carlo sampling places a hydroxyl on Cu B down to a pH below 4 and a water on heme a 3 beyond pH 11.…”

“…A different approach involves the use of heme-peptide complexes (called microperoxidases) obtained from the proteolysis of horse heart cytochrome c [20,21]. However, while bearing the advantages of good water solubility and reduced tendency to aggregate, which are helpful for the comparison with the proteins, microperoxidases suffer the problem of containing a different type of heme (heme c) in their structure.…”
