AbstrakBackground: Oligonol ® (OLG) is raw materials for functional foods or cosmetics which contains low-molecular-weight polyphenols derived from lychee fruit (Litchi chinensis Sonn.). It has been reported to many functions such as antiinflammatory and anti-oxidants. Aim: In this study, with the aim of exploring new functionalities of OLG on the scalp and hair growth, we investigate i ts effect on human dermal papilla cells (HDPC) by comparing it with adenosine and minoxidil at the genetic level. Method: OLG, adenosine, and minoxidil were applied to HDPC culture lines for 24 h, after which VEGF, FGF-7, WNT5a, and WNT10a mRNA expression was measured by real-time PCR analysis. Additionally, using DNA microarrays, we investigated their effects on 205 inflammation-related genes. Result: The FGF-7, and WNT10a mRNA expression were observed in HDPC added with OLG. The results of the DNA microarray analysis showed that the expression of 11 genes was suppressed by OLG. These functions of OLG on HDPC were not the same as adenosine and minoxidil. Conclusions: OLG may affect the function of HDPC by regulating the expression of genes related to cell proliferation and inflammation.

BackgroundLL-37 is a naturally occurring antimicrobial peptide found in the wound bed and assists wound repair. No published study has characterized the role of LL-37 in the function(s) of human mesenchymal stem cells (MSCs). This study investigated the functions of adipose-derived stromal/stem cells (ASCs) activated by LL-37 by performing both in vitro assays with cultured cells and in vivo assays with C57BL/6 mice with hair loss.MethodsHuman ASCs were isolated from healthy donors with written informed consent. To examine the effects of LL-37 on ASC function, cell proliferation and migration were measured by a cell counting kit (CCK-8) and a Transwell migration assay. Early growth response 1 (EGR1) mRNA expression was determined by microarray and real-time PCR analyses. The protein levels of EGR1 and regenerative factors were analyzed by specific enzyme-linked immunosorbent assays and western blotting.ResultsLL-37 treatment enhanced the proliferation and migration of human ASCs expressing formyl peptide receptor like-1. Microarray and real-time PCR data showed that EGR1 expression was rapidly and significantly increased by LL-37 treatment. LL-37 treatment also enhanced the production of EGR1. Moreover, small interfering RNA-mediated knockdown of EGR1 inhibited LL-37-enhanced ASC proliferation and migration. Activation of mitogen-activated protein kinases (MAPKs) was essential not only for LL-37-enhanced ASC proliferation and migration but also EGR1 expression; treatment with a specific inhibitor of extracellular signal-regulated kinase, p38, or c-Jun N-terminal kinase blocked the stimulatory effect of LL-37. EGR1 has a strong paracrine capability and can influence angiogenic factors in ASCs; therefore, we evaluated the secretion levels of vascular endothelial growth factor, thymosin beta-4, monocyte chemoattractant protein-1, and stromal cell-derived factor-1. LL-37 treatment increased the secretion of these regenerative factors. Moreover, treatment with the conditioned medium of ASCs pre-activated with LL-37 strongly promoted hair growth in vivo.ConclusionsThese findings show that LL-37 increases EGR1 expression and MAPK activation, and that preconditioning of ASCs with LL-37 has a strong potential to promote hair growth in vivo. Role of increased vascular permeability in chemotherapy‐induced alopecia: In vivo imaging of the hair follicular microenvironment in mice

Chemotherapy‐induced alopecia is one of the most difficult adverse events of cancer treatment for patients. However, it is still unknown why anticancer drugs cause hair loss. We aimed to clarify the mechanism of chemotherapy‐induced alopecia in mice using an in vivo imaging technique with a two‐photon microscope, which enables observation of the deep reaction in the living body in real time. In this study, ICR mice were injected intraperitoneally with cyclophosphamide (120 µg/g). Changes in the hair bulb morphology, subcutaneous vessel permeability, and vessel density were evaluated by two‐photon microscopy and conventional methods. In order to determine whether there is a causal relationship between vascular permeability and hair loss, we combined cyclophosphamide (50 µg/g) with subcutaneous histamine. We found that the hair bulbs became smaller, blood vessels around the hair follicle decreased in diameter, and vascular permeability was confirmed by two‐photon microscopy and conventional examination at 24 h after cyclophosphamide injection. Apoptosis occurred in vascular endothelial cells around the hair follicle. Additionally, hair loss was exacerbated by temporarily enhancing vascular permeability with histamine. In conclusion, cyclophosphamide caused a decrease in vascular density and an increase in vascular permeability, therefore increased vascular permeability might be one of the causes of chemotherapy‐induced alopecia.
