Helicobacter hepaticus infection in BALB/c mice abolishes subunit-vaccine-induced protection against M. tuberculosis

Abstract: HighlightsNeonatal Hh infection of mice upregulates colonic IL10 message.Neonatal Hh infection reduces lung immune responses after immunisation with Ad85A.Protection against Mtb challenge induced by Ad85A is abolished in Hh infected mice.IL10R blockade reverses the effects of Hh infection on Ad85A induced protection.Addition of Hh to the microbiota abolishes protection induced by a subunit vaccine.

“…43 Interestingly, these CIP effects were not influenced by the microbiome as the same effect was seen in germ-free mice. 43 While the microbiome and its metabolites can enhance or dysregulate the immune system, 44,45 CIP directly decreases immune strength and causes dysbiosis, while its metabolites enhance phagocytosis (macrophages) and reduce antimicrobial action. 42,43 These complex relationships require further study.…”

“…Cells were diluted to a final density of 1 ϫ 10 6 cells ml Ϫ1 in cRPMI, and cells were dispensed in propylene tubes for fluorescence-activated cell sorting (FACS) staining. Following removal of epithelial cells and collagenase VIII/DNase I (Roche) digestion of the tissue, lamina propria mononuclear cells were isolated as described previously (29). At the end of the process, cells were counted using the Scepter cell counter (Millipore) and diluted to a final concentration of 1 ϫ 10 6 cells ml Ϫ1 in cRPMI in propylene FACS staining tubes.…”

“…Two case‐contact studies in Gambia and Pakistan revealed association of the commensal opportunist, Helicobacter ( H.) pylori with protection to active tuberculosis, which was confirmed in experimentally infected cynomolgus macaques . In contrast, H. hepaticus conferred increased susceptibility of Balb/c mice to experimental tuberculosis . Dysbiosis in mice induced by antibiotic treatment promoted M. tuberculosis growth .…”

“…suggested that bacterial LPS augments liver‐toxicity of isoniazid . Vaccination efficacy to tuberculosis can also be influenced by commensals as shown for Helicobacter hepaticus in Balb/c mice and Bifidobacterium in infants .…”
