Heat Shock Protein 70 Expression Is Spatially Distributed in Human Placenta and Selectively Upregulated during Labor and Preeclampsia

Abstract: Placental oxidative stress is a feature of both human labor and the pregnancy syndrome preeclampsia. Heat shock proteins (HSPs) can be induced in cells as a protective mechanism to cope with cellular stress. We hypothesized that HSP 70 would increase during labor and preeclampsia and that expression would vary in different placental zones. Samples were obtained from 12 sites within each placenta: 4 equally spaced apart pieces were sampled from the inner, middle and outer placental regions. Non-labor, labor and… Show more

“…Tissues was homogenised in the presence of protease inhibitors as described previously [12]. Placenta homogenates were spun at 5000 g for 10 minutes at 4°C to remove debris then supernatants were collected and divided into aliquots and stored at −70°C.…”

“…We previously confirmed that scanning densitometry provided similar results to other quantitative methods [16]. Care was taken to ensure that the bands which were scanned fell on the linear range of protein loading as described previously [16]. Since several gels were run, the same internal loading control (one of the samples) was added to each gel and bands were normalised against this value.…”
