Heat shock protein 47 effects on hepatic stellate cell-associated receptors in hepatic fibrosis of Schistosoma japonicum-infected mice

Abstract: The study aimed to explore the regulation of heat shock protein 47 (HSP47) on expressions of receptors associated with hepatic stellate cell (HSC) in liver fibrosis mouse models induced by Schistosoma japonicum (S. japonicum). Mouse fibroblasts (NIH/3T3) were transfected with HSP47 shRNA plasmid by lipofectamine transfection, and experimental fibrosis in HSCs was studied in S. japonicum mouse models treated with HSP47 shRNA in vivo. HSP47 expression was assessed using Western blot and real-time PCR. Flow cytom… Show more

“…Macrophage polarization is related to parasitic infections. Macrophage polarization has been examined in mice with infection by schistosoma [ 19 ] and has also been reported in other parasitic diseases induced by toxoplasma gondii [ 20 ] and Clonorchis sinensis [ 21 ]. Macrophage polarization is associated with alveolar echinococcosis infection [ 22 ].…”

“…BMS‐986263/ND‐L02‐s0201 is a cationic LNP with surface conjugated retinoid moieties encapsulating a chemically modified siRNA oligonucleotide that targets heat shock protein 47 (HSP47), a collagen‐specific chaperone that is expressed in hepatic stellate cells. ( 88 ) As the chaperone of collagen, HSP47 regulates collagen synthesis, secretion, and maturation, which are associated with the development of liver fibrosis. ( 89 ) The LNP platform represents the leading formulation platform for effective delivery of therapeutic oligonucleotides to the target site.…”

“…HSP47 is presented in the endoplasmic reticulum and is crucial in controlling collagen synthesis. HSP47 is implicated in fibrotic disorders such as scleroderma, renal interstitial fibrosis, peritoneal fibrosis, cardiac fibrosis, intestinal fibrosis, keloid fibrosis, and pulmonary fibrosis by encouraging the buildup of collagen [ 26 ].…”
