Heart Failure-Induced Diaphragm Myopathy

Abstract: Background: Intracellular signaling pathways involved in skeletal myosin heavy chain (MyHC) isoform alterations during heart failure (HF) are not completely understood. We tested the hypothesis that diaphragm expression of mitogen-activated protein kinases (MAPK) and myogenic regulatory factors is changed in rats with myocardial infarction (MI) induced HF. Methods: Six months after MI rats were subjected to transthoracic echocardiography. After euthanasia, infarcted rats were subdivided in MI/HF- group (with n… Show more

“…All samples had 260/230 nm and 260/280 nm absorbance ratios higher than 1.8. RNA integrity was verified by electrophoresis on 1 % agarose gel [23]. RNA was reverse transcribed using High Capacity cDNA Reverse Transcription kit (Applied Biosystems, CA, USA) as recommended by the manufacturer.…”

“…Echocardiographic evaluation was performed using a commercially available echocardiograph (General Electric Medical Systems, Vivid S6, Tirat Carmel, Israel) equipped with a 5 - 11.5 MHz multifrequency probe [30,31,32]. Rats were anesthetized by intramuscular injection of a mixture of ketamine (50 mg/kg) and xylazine (0.5 mg/kg).…”
