Headache‐type adverse effects of NO donors: vasodilation and beyond

Abstract: Although nitrate therapy, used in the treatment of cardiovascular disorders, is frequently associated with side-effects, mainly headaches, the summaries of product characteristics of nitrate-containing medicines do not report detailed description of headaches and even do not highlight the possibility of nitrate-induced migraine. Two different types of nitrate-induced headaches have been described: (i) immediate headaches that develop within the first hour of the application, are mild or medium severity without… Show more

“…Most of the potentially severe side effects of glyceryl trinitrate overdose are related to its systemic vasodilator effect, which can cause hypotension and headaches (59,60). We therefore used blood pressure as the indicator for glyceryl trinitrate in vivo activity and to assess severe side effects.…”

“…Trigeminovascular system activation seems to be the key point for the initiation of migraine attacks. Therefore, NO may be involved in the initiation of a series of alterations, such as changes in the functions of ion channels, or calcitonin gene-related peptide (CGRP) or glutamate release that may result in migraine type headaches (14,15). CGRP is a significant molecule in this respect.…”

“…Mindkét betegség hátterében alapvető patofi ziológiai tényezők, például a fájdalomszabályozás, a peptiderg, a szerotonerg és a GABAerg rendszer vagy az ioncsatornák működésének a megváltozása áll [24]. Az akut és profi laktikus kezelésre is alkalmazott kalciumantagonista fl unarizin a betegek mintegy 8%-ában olyan súlyos depressziót okozott, ami a kezelés abbahagyását tette szükségessé [25], ezért csak második vonalbeli szernek alkalmazzák.…”
