HCV-coinfection is related to an increased HIV-1 reservoir size in cART-treated HIV patients: a cross-sectional study

Abstract: In HIV-1/HCV-coinfected patients, chronic HCV infection leads to an increased T-lymphocyte immune activation compared to HIV-monoinfected patients, thereby likely contributing to increase HIV-1 reservoir that is the major barrier for its eradication. Our objective was to evaluate the influence of HCV coinfection in HIV-1 viral reservoir size in resting (r) CD4+ T-cells (CD25-CD69-HLADR-). Multicenter cross-sectional study of 97 cART-treated HIV-1 patients, including 36 patients with HIV and HCV-chronic co-infe… Show more

“…Although the combined antiretroviral therapy (ART) can successfully suppress HIV viremia and delays the progression of disease [ 1 ], the chronic infection requires lifetime treatment due to the viral persistence in latent reservoirs [ 2 , 3 , 4 ]. Importantly, a significant percentage of HIV-infected individuals has hepatitis C virus (HCV) co-infection [ 5 ] resulting in increased HIV reservoir size [ 6 ]. The advent of ART has improved survival of HIV-infected adults and children leading to chronic illnesses such as different pulmonary diseases [ 7 , 8 ].…”

“…Fifty milliliters of blood samples were taken from all study patients. Briefly, fresh PBMCs were obtained and rCD4+ T cells were purified by negative selection with EasySep™ Human Resting CD4+ T-Cell Isolation Kit (Stemcell Technologies, Vancouver, Canada), as described previously by our group [ 9 ]. The purity of CD4+CD25-CD69-HLA/DR-T cells was assessed by flow cytometry as >99%.…”

“…Values of 2-LTR circle DNA molecules in rCD4 T+ cells and rCD4 T- PBMCs were subtracted from the integrated HIV-DNA value. Information regarding the methodology and procedures involved in the assessment of proviral DNA has been previously described by our group elsewhere [ 9 ].…”
