Hazara Nairovirus Requires COPI Components in both Arf1-Dependent and Arf1-Independent Stages of Its Replication Cycle

Abstract: Hazara nairovirus (HAZV) is an enveloped tri-segmented negative strand RNA virus classified within the Nairoviridae family of the Bunyavirales order, and a member of the same subtype as Crimean-Congo hemorrhagic fever virus, responsible for fatal human disease. Nairoviral subversion of cellular trafficking pathways to permit viral entry, gene expression, assembly and egress is poorly understood. Here, we generated a recombinant HAZV expressing eGFP and used live-cell fluorescent imaging to screen an siRNA libr… Show more

“…We also tested the dependency to LDL-R of Hazara virus (HAZV), another member of the genus Orthonairovirus , using a GFP-expressing recombinant virus 24 produced in Huh-7.5 cells. Interestingly, the blocking of LDL-R at the surface of Huh-7.5 cells did not impair HAZV infection as assessed by flow cytometry (Fig.…”

“…Previous studies reported that the GBF1-ARF1-COP I axis was required for the replication of various viruses, including dengue virus (DENV) ( 13 ), HCV ( 26 ), and chikungunya virus (CHIKV) ( 27 ), which is consistent with our findings. However, Hazara nairovirus (HAZV) also utilizes COP I in an ARF1-independent manner to promote genome replication, possibly because the transport function of COP I-coated vesicles was not required for this step ( 28 ).…”

“…We have attempted to isolate MHMV with Vero E6, SW-13, and BHK-21 cell lines that have been used for isolation of NSD genogroup viruses (CCHFV, HAZV, and TFLV; Shimada et al, 2016 ; Fuller et al, 2020 ; Dai et al, 2021 ). However, neither CPE nor viral RNA has been detected across several passages in cell cultures.…”
