Harnessing the Natural Healing Power of Colostrum: Bovine Milk‐Derived Extracellular Vesicles from Colostrum Facilitating the Transition from Inflammation to Tissue Regeneration for Accelerating Cutaneous Wound Healing

Abstract: As wound healing is an extremely complicated process, consisting of a cascade of interlocking biological events, successful wound healing requires a multifaceted approach to support appropriate and rapid transitions from the inflammatory to proliferative and remodeling phases. In this regard, here the potential use of bovine milk extracellular vesicles (EVs) to enhance wound healing is investigated. The results show that milk EVs promote fibroblast proliferation, migration, and endothelial tube formation. In p… Show more

“…In particular, the enhanced migration result was found in the Col M-exo-treated group (Figure S2A). Consistent with our previous study that Col M-exo was rich in collagen and elastin deposition proteins and cell proliferation and migration-promoting proteins compared to Mat M-exo [49], the fibroblast migration ability was improved in the Col M-exo-treated group.…”

“…In particular, the enhanced migration result wa found in the Col M-exo-treated group (Figure S2A). Consistent with our previous study that Col M-exo was rich in collagen and elastin deposition proteins and cell proliferation and migration-promoting proteins compared to Mat M-exo [49], the fibroblast migration ability was improved in the Col M-exo-treated group. Taken together, these findings suggest that Col M-exo can effectively promote ECM construction by improving survival of skin fibroblasts and collagen production under UV irradiation, offering strength and elasticity to the skin.…”

“…Oral ingestion of bovine colostrum particularly showed no side effects in clinical research even on repeated application of high doses, except lactose intolerance ( Struff and Sprotte, 2008 ). Moreover, milk-derived exosomes have a very high yield compared to animal cell-derived exosomes, which has economic advantages for drug commercialization ( Kim et al, 2021 ).…”

“…Bovine colosEVs also showed shared effects with the mEVs that were mediated by the inflammatory immune regulation, thereby improving colitis symptoms in a mouse model [ 197 ]. The capacity to reduce inflammation could be useful also in wound healing, for which colosEVs shifted the inflammatory process to fibroblast proliferation, migration, and endothelial tube formation, thereby accelerating the wound recovery [ 198 ]. Like bovine mEVs, cow colosEVs were able to shape the microbiota; this was correlated with bone remodelling and partial recovery in an osteoporosis mouse model [ 199 ].…”
