Haplopine Ameliorates 2,4-Dinitrochlorobenzene-Induced Atopic Dermatitis-Like Skin Lesions in Mice and TNF-α/IFN-γ-Induced Inflammation in Human Keratinocyte

Abstract: This study aimed to investigate the anti-inflammatory, antioxidant, and anti-atopic dermatitis (AD) effects of haplopine, which is one of the active components in D. dasycarpus. Haplopine (12.5 and 25 mM) inhibited the mRNA expressions of inflammatory cytokines IL-6, TSLP, GM-CSF, and G-CSF and the protein expressions of IL-6 and GM-CSF in TNF-α/INF-γ-stimulated HaCaT cells. In H2O2-induced Jukat T cells, haplopine (25 and 50 mM) suppressed the productions of proinflammatory cytokines (IL-4, IL-13, and COX-2) … Show more

“…More related antioxidants are still being studied. For example, it has recently been found that Haplopine, a biologically active component of plants, increases the expression of SOD, CAT, HO-1 in a concentration-dependent manner in vitro and has excellent therapeutic potential in AD mouse models ( Kim et al, 2021 ). Many inorganic substances have strong antioxidant properties, such as the previously mentioned H 2 S (one of the RSS).…”

“…The expression of HO-1 was increased when the skin is exposed to environmental stimuli such as H 2 O 2 induced oxidative stress or in ammatory factors to give play to cytoprotective effects of anti-in ammatory [12], antioxidant [13], anti-apoptotic [14] and maintenance of epithelial cell integrity [15]. HO-1 attenuates the development of atopic dermatitis-like lesions in mice [16][17][18], suggesting HO-1 may play a certain role in the development of AD, but the mechanisms of its action involved are not fully understood.…”

“…To date, there is no report on the production of these growth factors in the infection of keratinocytes by Sporothix spp. However, the production of GM-CSF and G-CSF has been reported in keratinocytes treated with dinitrochlorobenzene, and they have been attributed a pro-inflammatory effect [ 65 ], which could be contributing to the pathogenesis of Sporotrichosis.…”
