SummaryThe effect of nitrogen (N) nutrition upon the accumulation of seed storage protein has been studied in wildtype and opaqoe-2 mutant (02) maize endosperms grown in vitro, for 5 days, on a solid medium containing different amounts of N in the form of inorganic or organic compounds. As reference, the accumulation of zein proteins in endosperms grown to maturity in vivo under field conditions was also examined. The developmental stage up to 14-19 days after pollination (DAP) defines, both in vivo and in vitro, a phase of growth during which the major zein classes of 20 and 22 kDa are synthesized both in wild-type and o2endo-sperms. In vitro, the corresponding transcripts of the genes coding the 20 and 22 kDa zeins, and of the zein synthesis associated gene b-32, are also present. Under in vitro conditions, the level of expression of the zein synthesizing system, as monitored by zein accumulation, is dependent on the level and form of N supplied in the medium. In vivo, at stages of development from 19 DAP to maturity, the synthesis of the 22 kDa zein class and of the b-32 protein is under control of the 02 transcriptional activator. It is hypothesized that at early stages of endosperm development (between 9 and 14-19 DAP) a 'metabolic control', based on N availability, is operative, possibly based on a regulatory factor different from 02.

Camelina [Camelina sativa (L.) Crantz], a member of the Brassicaceae family, has a unique oil profile that has potential both for biofuels and as a food crop. It is essential to have a doubled haploidy protocol in order to enhance breeding of this crop for prairie conditions as well as improve the yield and quality characteristics, Microsporederived embryos have been produced from Camelina sativa. Buds 1 -3 mm in length were selected for culture.The microspores were isolated and purified in full-strength B5 extraction medium and cultured in NLN medium with 12.5% sucrose and 12.5% polyethylene glycol 4000 (PEG) without glutamine, at a density of 10,000 microspores per mL. Glutamine was added to the cultures 72 h after extraction to give a final concentration of 0.8 g/L. The microspore cultures were maintained at 24°C in the dark. After 28 days embryos were observed and these were regenerated to plants and selfed seed was produced. The highest embryogenic frequency achieved was 38 microspore-derived embryos from 100,000 microspores.

ABSTRACT:Two released noug varieties, Shambu and Fogera, were used for this experiment.Initially, two experiments were conducted to identify the appropriate morphological stage of capitula (buds) for harvesting maximum number of responsive anthers and to study the optimum cold pretreatment duration for callus induction from the anthers. Three culture media, MS, BB 5 and NN, were employed for callus induction. MS medium, with nine different combinations of KN and IAA, was used for shoot regeneration study. Three different concentrations of IBA and growth regulator-free MS medium were used for the rooting experiment. The results showed that capitula, fully or slightly covered by sepal having whitish-green or greenish-yellow anthers, were the optimum stages of harvesting capitula; callus was efficiently induced when the capitula were pretreated at 4 C for 24 hours. The types of calli induced significantly (P≤0.05) varied among the three media. Embryogenic calli were mainly produced in NN and B 5 °B media, while varying in texture and colour. High percentage of embryogenic calli (80%) was induced on NN medium from the variety Shambu, followed by BB 5 medium showing 40% inductions for both varieties. Among nine different KN and IAA combinations, 2 mg/l KN combined with 1 mg/l IAA, was the most efficient for shoot regeneration. It was also found that 0.5 mg/l IBA was optimum for root induction. In the growth room as well as in the glasshouse, the survival rate of regenerants was generally better for the variety Shambu than for Fogera.
