Haploid Plants from Pollen Grains

Abstract: A method is presented by which hundreds of haploid plants of various species of Nicotiana can be raised from pollen grains. Stamens should be excised when pollen grains have been individualized, but are still uninucleate and free of starch. When grown in vitro on a relatively simple medium, some pollen grains proliferate into embryo-liké, structures which develop in stages similar to those of zygotic embryos. The plantlets mature and flower profusely, but do not set seed.

“…Media were prepared as described in Table 1. with Nitsch and Nitsch (1969) salts or the same salts lacking ammonium nitrate, with the addition of 0.4 mg 1 -l thiamine-HCl, 100 mg 1 -l inositol, 30 g I-' sucrose, and different concentrations of L-asparagine. The salt solutions were adjusted to pH 6.0 and sterilized with the agar in the autoclave (120"C, 1.2 kg crn-*, 20 min), while the organic nutrients were filter-sterilized and added later.…”

“…N6 (Chu 1978) and KFWC (Kuhlmann and Foroughi-Wehr 1989). There was minimal response from NN (Nitsch and Nitsch 1969) (data not shown). Callus and limited embryo development was also observed in NLN.…”

“…After cold-pretreatment using the optimum time duration (24 hours), anthers of the varieties Shambu and Fogera taken from Stage 1 capitula , were cultured on three different culture media, MS (Murashige and Skoog, 1962), B 5 (Gamborg et al, 1968) and NN (Nitsch and Nitsch, 1969). The basal media contained 2% sucrose, all supplemented with 2 mg/l NAA plus 0.3 mg/l KN and solidified with 0.8% agar.…”
