Habenula as a Possible Target for Treatment-Resistant Depression Phenotype in Wistar Kyoto Rats

Abstract: The mechanisms of treatment-resistant depression (TRD) are not clear and are difficult to study. An animal model resembling human TRD is the Wistar Kyoto rat strain. In the present study, we focused on selecting miRNAs that differentiate rats of the WKY strain from Wistar Han (WIS) rats in two divisions of the habenula, the lateral and medial (LHb and MHb, respectively). Based on our preliminary study and literature survey, we identified 32 miRNAs that could be potentially regulated in the habenula. Six miRNAs… Show more

“…In addition, the WKY rats show hypoactivity in open field tests, high anxiety (Dugovic et al, 2000) and depressive behaviour (Pare, 1992(Pare, , 1994. Of particular relevance to the present study, there are significant differences in dopamine transporter expression in the nucleus accumbens and VTA of WKY rats compared to Wistar and SD strains (Jiao et al, 2003) and differences in dopamine receptor expression (Korlatowicz et al, 2023;Morganstern & Tejani-Butt, 2010;Yaroslavsky et al, 2006). These behavioural and neurochemical characteristics make WKY unsuitable as a reference strain in the current study.…”

“…In addition, downregulation of several serotonergic receptor genes was reported in stressed rats compared to non-stressed controls (73). Serotonergic dysfunction has also been reported in the Hb of the Wistar Kyoto rat, an endogenous model of treatment resistant depression (89), with downregulation of the serotonergic receptor genes Htr7 and Htr2a in both the MHb and LHb and increased expression of Htr4 in the LHb but not MHb (80). Similarly, a reduction in 5-HTR7 receptor expression was isolated to the LHb (80).…”

“…Previous studies comparing the two strains of animals have shown that they differ significantly in terms of biochemistry [ 9 ]. Recently, we have demonstrated that several important miRNAs are expressed in the habenula, which differentiated WKY rats from WIS rats and were correlated with changes in the expression of target transcripts [ 41 ]. However, it turns out that the WKY strain is more complicated.…”
