H‐Ras increases urokinase expression and cell invasion in genetically modified human astrocytes through Ras/Raf/MEK signaling pathway

Abstract: Previous study reported that the activation of Ras pathway cooperated with E6/E7-mediated inactivation of p53/pRb to transform immortalized normal human astrocytes (NHA/hTERT) into intracranial tumors strongly resembling human astrocytomas. The mechanism of how H-Ras contributes to astrocytoma formation is unclear. Using genetically modified NHA cells (E6/E7/ hTERT and E6/E7/hTERT/Ras cells) as models, we investigated the mechanism of Ras-induced tumorigenesis. The overexpression of constitutively active H-Ras… Show more

“…An increasing number of studies demonstrated that MMP-2 and MMP-9 have important roles in GBM angiogenesis and can be regulated by multiple pathways (23)(24)(25). The present study examined MMP-2 and MMP-9 expression to explore the association between the hedgehog pathway and cell invasion and migration.…”

“…Recent study [25] showed that H-Ras increased urokinase (uPA) expression and contributed to increased invasiveness of astrocytomas. Intranasal administration of perillyl alcohol, a Ras inhibitor, to patients with recurrent gliomas improved the overall survival of patients included in the present cohort.…”

“…Equal amounts of proteins (30 g) were loaded in each lane for matrix metalloproteinase (MMP)-2 and -9 gelatinolytic activity detection by use of gelatin zymography (36 -38). Urokinase plasminogen activator and tPA activities were detected by plasminogen-fibrinogen zymography (35).…”
