GWAS loci associated with Chagas cardiomyopathy influences DNA methylation levels

Abstract: A recent genome-wide association study (GWAS) identified a locus in chromosome 11 associated with the chronic cardiac form of Chagas disease. Here we aimed to elucidate the potential functional mechanism underlying this genetic association by analyzing the correlation among single nucleotide polymorphisms (SNPs) and DNA methylation (DNAm) levels as cis methylation quantitative trait loci (cis-mQTL) within this region. A total of 2,611 SNPs were tested against 2,647 DNAm sites, in a subset of 37 chronic Chagas … Show more

“…More precisely, all the previously described biological process seems to be linked to the immune response, and notably to the Th1 response, including IFN-γ and RUNX3. These results, combined with those obtained in previous analyses of CCC ( 9 , 78 ), confirm the importance of DNA methylation in the development of CCC.…”

“…It is particularly applied to understanding the genetic basis of complex traits and diseases. Furthermore, it has also been successfully used to study mQTLs (methylation QTLs) and thus relate changes in the methylation of CpG sites, usually in gene regulatory regions, to polymorphic variations [38] .…”

“…We also note the few members of the set of phospholipase A and acyltransferase paralogs (PLAAT1, 2 and 4) that lack a CVD-related bibliography (see Table 2), while PLAAT3 (a.k.a. PLA2G16) has been found in a GWAS study associated with Chagas cardiomyopathy [37] and was the most downregulated gene in calf muscle of patients diagnosed with peripheral artery disease (PAD) [38].…”
