Abstract: We assume that detoxification enzymes are functionally redundant and that only the simultaneous deficiency of several detoxification enzymes increases the risk for oral cancer.

“…5,44,81,82,99,100 Hospital patients were used as controls in 16 studies. 5,44,71,72,78,[81][82][83][84][85]88,92,93,[97][98][99] The number of cases in the studies included in the meta-analysis for GSTM1 deletion varied from 21 to 451 patients. All studies undertaken in Europe included Ͻ150 cases, with two of these having Ͻ50 cases.…”

The frequency of the homozygote genotype for the variant allele in the controls was absent or very low in Europe (0 -6%) whereas the heterozygous genotype was very rare (6 -9.3%).…”

“…Metabolism of this carcinogen involves a balance between the activation steps mediated by the epoxide hydrolase and the cytochrome system and the detoxification steps, involving GSTM1, that inhibit the activity of the DNA binding intermediates and catalyze the conversion of the reactive electrophiles to inactive, water-soluble conjugates that can be easily removed [13]. Even though frequencies of GSTM1 null genotypes vary among different ethnic groups, in white Caucasians, it is deleted in approximately 50% of the population [2433]. …”

“…The homozygous deletion (0/0), or null genotype, which leads to copy number variations (CNVs) at either the GSTM1 or the GSTT1 locus, resulted in loss of enzyme function. Originally, it was hypothesized that this locus was associated with the susceptibility to autoimmune conditions, such as lupus erythematosus and various forms of cancer, including oral, gastric, colorectal, prostate, bladder, and hepatic cancer and renal cell carcinoma (Economopoulos and Sergentanis 2010 ;Gronau et al 2003 ;Hayes and Pulford 1995 ;Simic et al 2009 ;Zhang et al 2010Zhang et al , 2011.…”
