SummaryCyclin-dependent kinase (CDK) inhibitor genes encode low molecular weight proteins which have important functions in cell cycle regulation, development and perhaps also in tumorigenesis. The first plant CDK inhibitor gene ICK1 was recently identified from Arabidopsis thaliana. Although the C-terminal domain of ICK1 contained an important consensus sequence with the mammalian CDK inhibitor p27 Kip1 , the remainder of the deduced ICK1 sequence showed little similarity to any known CDK inhibitors. In vitro assays showed that recombinant ICK1 exhibited unique kinase inhibitory properties. In the present study we characterized ICK1 in terms of its gene structure, its interaction with both A. thaliana Cdc2a and CycD3, and its induction by the plant growth regulator, abscisic acid (ABA). ICK1 was expressed at a relatively low level in the tissues surveyed. However, ICK1 was induced by ABA, and along with ICK1 induction there was a decrease in Cdc2-like histone H1 kinase activity. These results suggest a molecular mechanism by which plant cell division might be inhibited by ABA. ICK1 clones were also identified from independent yeast two-hybrid screens using the CycD3 construct. The implication that ICK1 protein could interact with both Cdc2a and CycD3 was confirmed by in vitro binding assays. Furthermore, deletion analysis indicated that different regions of ICK1 are required for the interactions with Cdc2a and CycD3. These results provide a mechanistic basis for understanding the role of CDK inhibitors in cell cycle regulation in plant cells.

It is well established that auxins play a central role in the determination of rooting capacity, which is essential for vegetative propagation. Recent studies with apple trees have pointed to significant effects of auxin stability, wound related phenolics and ethylene production in the control of adventitious rooting. In the present study, a comparative analysis of the adventitious rooting of microcuttings of Eucalyptus saligna (easy-to-root species) and Eucalyptus globulus (difficult-to-root species) was carried out with different types of auxins, light intensities, presence or absence of apical meristem, different concentrations of phenolic compounds and presence or absence of an ethylene action inhibitor. Parameters evaluated were the percent rooting, number of roots per rooted cutting, length of longest root and mean rooting time. Results showed that auxins of intermediate stability are more favorable to rooting (particularly for the recalcitrant species), higher light intensities in the presence of exogenous auxins promote the rooting response, the absence of meristematic apex or externally supplied phenolics are not limiting for the rooting induced by exogenous auxins, and ethylene appears to play a minor role in the development of adventitious roots in microcuttings of Eucalyptus, indicating that the rhizogenic response results from direct effect of auxins.

Alstonia scholaris (L.) R. Br. is an indigenous plant species in Indonesia which has been use for multiple purposes. This plant tolerant to poor nutrient and alkaline soil, therefore it is potential to explore as a revegetation plant. The study was conducted to identify effect of IBA concentrations on rooting ability of A. scholaris shoot cutting. Plant materials (shoot cutting) were collected from trubusan in Mandeh Area, Tarusan, West Sumatra. This study used Completely Randomized Design (CRD). The treatment was IBA concentrations (0 ppm, 50 ppm, 100 ppm, 150 ppm and 200 ppm). The result showed that IBA concentrations was significant different (P ? 0,05) for length, fresh weight and dry weight of root. The application of 150 ppm IBA was the best concentration to enhance the rooting ability of A. scholaris shoot cuttings.
