Growth Rates of Protein Crystals

Abstract: Protein crystallization is important for structural biology. The rate at which a protein crystallizes is often the bottleneck in determining the protein’s structure. Here, we give a physical model for the growth rates of protein crystals. Most materials crystallize faster under stronger growth conditions, however, protein crystallization slows down under the strongest conditions. Proteins require a crystallization slot of ’just right’ conditions. Our model provides an explanation. Unlike simpler materials, pro… Show more

“…Because for the vast majority of proteins no structural information is available at all, the former is a genuine difficulty. The latter is also important, because evidence suggests that non-crystallographic contacts may play a kinetic role in protein crystal assembly [147] and may even render metastable crystal phases kinetically accessible [139].…”

“…Interestingly, it is well-known that the size and morphology of crystals can depend on various factors. Different researches have contributed to shed light on the critical parameters, such as precipitant concentration, buffer composition, pH, supersaturation and temperature [3,4], high or low supersaturation condition [5] or contaminant concentration [6]. Moreover, some scholars have successfully managed to crystallize lysozyme using solid flat surfaces or templates, observing that the nature of each template has a peculiar influence on lysozyme crystal nucleation and growth [5].…”
