Growth of Venezuelan Equine Encephalomyelitis Virus in Human Diploid Cell Strain WI-38

Abstract: We demonstrated that Venezuelan equine encephalomyelitis (VEE) virus replicated in and adapted rapidly to human diploid cell strain WI-38. Peak titers of approximately 109.8 mouse intracerebral 50% lethal doses were obtained at low passage levels in Eagles basal medium supplemented with calf serum. VEE virus replicated poorly in serum-free medium. Propagation of VEE virus was accompanied by the production of hemagglutinin and cytopathogenic effects.

“…Preparation of vaccines. Tissue culture virus suspensions were prepared in chick fibroblast monolayers, in Maitland-type chick embryo suspensions (MTCE) as described previously (15), and in monolayers of WI-38 cells (16). Virus suspensions were clarified by centrifuging at 700 X g in a PR-2 (I.E.C.)…”

“…Ten per cent or more of the animals were bled intracardially before vaccination, 21 days postvaccination, and 14 days postchallenge. Serum samples were assayed for hemagglutinatinginhibiting (HI; reference 14) and serum-neutralizing (SN) antibodies as previously described (15). SN indexes are expressed as log10 ; HI titers are expressed as the reciprocal.…”
