Growth/Differentiation Factor-10: A New Member of the Transforming Growth Factor-β Superfamily Related to Bone Morphogenetic Protein-3

Abstract: We have identified a new member of the transforming growth factor-beta (TGF-beta) superfamily, growth/differentiation factor-10 (GDF-10), which is highly related to bone morphogenetic protein-3 (BMP-3). The nucleotide sequence of GDF-10 encodes a predicted protein of 476 amino acids with a molecular weight of approximately 52,000. The GDF-10 polypeptide contains a potential signal sequence for secretion, a putative RXXR proteolytic processing site, and a carboxy-terminal domain with considerable homology to ot… Show more

“…It can induce endochondral bone formation (Wozney et al, 1988). The results of Cunningham et al (1995) in previous studies in mice suggest that GDF 10 may play multiple roles in regulating cell differentiation events, including those involved in skeletal morphogenesis. GDF 10 has been discovered in rat and human femur tissue (Hino et al, , 1999.…”

“…GDF10 was previously identified as a TGF-β ligand related to BMP-3 (14), although there is little information on its mechanism of action. Among several TGF-β family ligands, TGF-βs and activins signal primarily through activation of Smad2 and Smad3, whereas BMPs act through Smad1 and 5 (15,16).…”

“…Bone morphogenetic proteins (BMPs) and growth differentiation factors (GDFs) belong to the TGF-β superfamily (Hogan, 1996;Bhatia et al, 1999;Celeste et al, 1990;Cunningham et al, 1995;Storm et al, 1994;Dunn et al, 1997;Padgett et al, 1987). For the BMPs and GDFs only three type-I receptors and three type-II receptors have been identified, all of which are serine threonine kinases.…”
