Sepsis‐induced liver injury is recognized as a key problem in intensive care units. The gut microbiota has been touted as an important mediator of liver disease development; however, the precise roles of gut microbiota in regulating sepsis‐induced liver injury are unknown. Here, we aimed to investigate the role of the gut microbiota in sepsis‐induced liver injury and the underlying mechanism. Cecal ligation and puncture (CLP) was used to induce polymicrobial sepsis and related liver injury. Fecal microbiota transplantation (FMT) was used to validate the roles of gut microbiota in these pathologies. Metabolomics analysis was performed to characterize the metabolic profile differences between sepsis‐resistant (Res; survived to 7 days after CLP) and sepsis‐sensitive (Sen; moribund before or approximately 24 hours after CLP) mice. Mice gavaged with feces from Sen mice displayed more‐severe liver damage than did mice gavaged with feces from Res mice. The gut microbial metabolic profile between Sen and Res mice was different. In particular, the microbiota from Res mice generated more granisetron, a 5‐hydroxytryptamine 3 (5‐HT3) receptor antagonist, than the microbiota from Sen mice. Granisetron protected mice against CLP‐induced death and liver injury. Moreover, proinflammatory cytokine expression by macrophages after lipopolysaccharide (LPS) challenge was markedly reduced in the presence of granisetron. Both treatment with granisetron and genetic knockdown of the 5‐HT3A receptor in cells suppressed nuclear factor kappa B (NF‐кB) transactivation and phosphorylated p38 (p‐p38) accumulation in macrophages. Gut microbial granisetron levels showed a significantly negative correlation with plasma alanine aminotransferase (ALT)/aspartate aminotransferase (AST) levels in septic patients. Conclusion: Our study indicated that gut microbiota plays a key role in the sensitization of sepsis‐induced liver injury and associates granisetron as a hepatoprotective compound during sepsis development.

Background: Salmonella Enteritidis (SE) is one of the major foodborne zoonotic pathogens of worldwide importance which can induce activation of NLRC4 and NLRP3 inflammasomes during infection. Given that the inflammasomes play an essential role in resisting bacterial infection, Salmonella has evolved various strategies to regulate activation of the inflammasome, most of which largely remain unclear. Results: A transposon mutant library in SE strain C50336 was screened for the identification of the potential factors that regulate inflammasome activation. We found that T3SS-associated genes invC, prgH, and spaN were required for inflammasome activation in vitro. Interestingly, C50336 strains with deletion or overexpression of Dam were both defective in activation of caspase-1, secretion of IL-1β and phosphorylation of c-Jun N-terminal kinase (Jnk). Transcriptome sequencing (RNA-seq) results showed that most of the differentially expressed genes and enriched KEGG pathways between the C50336-VS-C50336Δdam and C50336-VS-C50336::dam groups overlapped, which includes multiple signaling pathways related to the inflammasome. C50336Δdam and C50336::dam were both found to be defective in suppressing the expression of several anti-inflammasome factors. Moreover, overexpression of Dam in macrophages by lentiviral infection could specifically enhance the activation of NLRP3 inflammasome independently via promoting the Jnk pathway. Conclusions: These data indicated that Dam was essential for modulating inflammasome activation during SE infection, there were complex and dynamic interplays between Dam and the inflammasome under different conditions. New insights were provided about the battle between SE and host innate immunological mechanisms.

Background: Most cases of gastric cancer (GC), which is one of the most common cancers in China, are diagnosed at an advanced stage. The first-line treatments for GC include 5-fluoropyrimidine (5-FU)-based chemotherapy. However, the cancer cells may develop 5-FU resistance. The inhibition of angiogenesis can be an alternative therapeutic strategy for GC. Recent studies have demonstrated that TIE2-expressing monocytes/macrophages (TEMs), a subtype of tumor-associated macrophages (TAMs), exhibit profound pro-angiogenic activity. The herbs used in traditional Chinese medicine can be a potential source of anti-cancer agents and can improve the therapeutic efficacy of chemotherapy drugs. In this study, the role of TEMs in the synergistic anti-cancer effects of Fu-Zheng-Jie-Du ointment (FZJD) and 5-FU was evaluated.Methods: The migration of TEMs was evaluated using the transwell assay. Tube formation assay and quantitative real-time polymerase chain reaction analysis were used to determine the pro-angiogenic activity of TEMs. Tumor-bearing mouse model and immunohistofluorescence stain were used to evaluate the anti-tumor effect of FZJD and the underlying mechanism.Results: FZJD inhibited the migration and pro-angiogenic activity of TEMs. Additionally, tumor growth in the mice co-treated with FZJD and 5-FU was lower than that in the mice treated with FZJD or 5-FU. Immunohistofluorescence stain revealed that the migration of TEMs was not inhibited upon treatment with FZJD alone or in combination with 5-FU. Moreover, treatment with FZJD did not decrease the endothelial cell population. In contrast, the treatment combination of FZJD and 5-FU markedly decreased the endothelial cell population.Conclusions: FZJD improves the anti-cancer efficacy of 5-FU through the inhibition of TEM-mediated angiogenesis.
