Groupers are important commercial fish in many parts of the world. Accurate identification is critical for effective conservation assessment and fisheries management. Genetic barcodes provide a simple and reproducible method for the identification of species even in the absence of taxonomic expertise. The generation of reference barcodes from properly identified specimens is an important first step in this direction. Here, 36 species belonging to the subfamily Epinephelinae (Family: Serranidae) were collected from landings on the west coast of India and Port Blair, Andaman, and partial nucleotide sequence data of the mitochondrial cytochrome C oxidase subunit I (COI) gene was generated. Barcodes for 13 species were developed from Indian waters for the first time. Analysis using the COI gene produced phylogenetic trees in concurrence with other multi-gene studies. Epinephelus fasciatus and E. areolatus were found to be a species complex, as hypothesized in other studies. The DNA barcodes developed in the study can be used for identifying species within Epinehelinae, where taxonomic ambiguity still exists. ARTICLE HISTORY

Objective. To assess the survival and growth of juvenile Pacific goliath grouper (Epinephelus quinquefasciatus) in captivity at low salinities. Materials and methods. We randomly selected twelve juvenile goliath groupers with an average standard length of 44.2 ± 6.1 cm and 1492 ± 476 g in body weight, and raised them for three months in the laboratory in waters of 32-26, 20, 10 and 5 parts per thousand of salinity. Results. All juvenile Pacific goliath groupers tolerated the initial gradual transfer from full strength seawater to salinities of 32-26, 20, 10, and 5 parts per thousand, which was done over a period of four days. All of them survived and grew in body weight and length during the course of the three-month experiment, in all the treatments of high and low salinity water. The body condition factor (K) for each fish was between 1.5 and 2.4. Conclusions. This is a first time trial that documents a randomized, controlled experiment demonstrating the ability of Pacific juvenile goliath grouper to gradually transfer from full strength seawater to water of lower salinity, and survive and grow well in these brackish waters.

ABSTRACT. Orange-spotted grouper (Epinephelus coioides) is one of the most important marine fish and has a high market value. The insulin-like growth factor type 1 receptor (IGF1R) is a component of the insulin-like growth factor signaling system, and demonstrates important roles during growth. Based on information from livestock, we used IGF1R as a candidate gene to survey single nucleotide polymorphisms. In the present study, the sequences of IGF1Ra and IGF1Rb from orange-spotted grouper were obtained from the genome sequences and their clustering in clades a and b, respectively, was confirmed by phylogenetic analysis. Fourteen critical amino acids underlying functional divergence were detected between the two clades, revealing the molecular basis of their functional differences. Nearly one-fourth (22 kbp) of the genomic sequence of IGF1Ra was sequenced in a mass cross population, and nucleotide diversity and linkage disequilibrium levels were investigated. Nucleotide diversity was 0.00328 for p and 0.00344 for q w . The half decay of the squared allele-frequency correlation was 10,835 base pairs. Comparatively, the relatively high level of linkage and the significant deviation from neutrality-based codons in IGR1R showed that this gene was under selection. A site (KR269824.1:g. 63762C>T), located in the sixth intron, was significantly associated with eyeball diameter (P = 1.39 x 10 -4 , Q-value: 2.33 x 10 -2 ), which accounted for 11.1% of phenotypic variance. These results highlight the important function of IGF1R in orange-spotted grouper and may be beneficial in the breeding of this species.
