Greater antitumor efficacy of paclitaxel administered before epirubicin in a mouse mammary carcinoma

Abstract: Combined treatment with paclitaxel and anthracyclines is increasingly being tested in clinical practice. Epirubicin is in general administered before paclitaxel. We have investigated, using a murine mammary adenocarcinoma, whether the efficacy and toxicity of this combination is influenced by treatment sequence, different time intervals and dose intensity. The tumor was transplanted into the right hind foot of C3D2F1 female mice. Paclitaxel was administered i.p. in doses ranging from 15 mg/kg to 75 mg/kg and e… Show more

“…Concentrations of chemotherapeutic drugs in our in vitro experiments approximate the maximal initial plasma concentrations ( c max ) observed in chemotherapy patients, and have been previously discussed [18,19]. Concentrations of agents administered to healthy mice are similar to doses administered in previous studies [20–22].…”

“…injection. 34 In a pilot study, a weekly i.p. injection of 20 mg/kg paclitaxel for 3 weeks led to frequent severe toxicities or death (four out of six mice), while a dose of 10 mg/kg of paclitaxel was found to be well tolerated.…”

“…Daily abdominal swelling, abdominal distension, regular activity, weight loss, muscle wasting, and any other signs or symptoms of toxicity were assessed. 23 , 24 , 25 The other two groups received RT in two fractions of 1 and 2 Gy each at 48‐h intervals. The animals were then anesthetized intravenously with xylazine/ketamine (10 mg/kg xylazine and 90 mg/kg ketamine).…”
