The Bonin Islands, comprising of the Mukojima, Chichijima, and Hahajima Islands, are known for their isolated and distinctive habitats, hosting a diverse array of endemic flora and fauna. In these islands, adaptive radiation has played a remarkable role in speciation, particularly evident in the Callicarpa genus that is represented by three species: Callicarpa parvifolia and Callicarpa glabra exclusive to the Chichijima Islands, and Callicarpa subpubescens, distributed across the entire Bonin Islands. Notably, C. subpubescens exhibits multiple ecotypes, differing in leaf hair density, flowering time, and tree size. In this study, we used double-digest restriction site–associated DNA sequencing to analyze species, ecotypes and geographical variations within Callicarpa in the Bonin Islands. We aimed to determine detailed phylogenetic relationships, investigate species and ecotype diversification patterns, estimate divergence times, and explore cryptic species using genetic and phenotypic data. Genetic analysis revealed that C. parvifolia and C. glabra formed a single, distinct genetic groups. Conversely, C. subpubescens showed seven genetic groups corresponding to different ecotypes and regions, with one ecotype derived from the hybridization of two others. Phylogenetic and population demography analyses, focusing on six Chichijima and Hahajima Islands–based species/ecotypes, indicated the divergence of an ecotype adapted to tall mesic forests approximately 170 kya, whereas the other five species/ecotypes diverged nearly simultaneously around 73–77 kya. Environmental changes during the glacial period likely contributed to this process of adaptive radiation. Moreover, leaf morphology, flowering time, and genetic analyses suggested the presence of two cryptic species within C. subpubescens.

Atomic absorption spectrometry is one of the effective methods for the determination of trace amounts of metal ions in water samples. However, many preconcentration techniques, such as solvent extraction 1 , coprecipitation 2 and ion exchange 3 , were combined with the atomic absorption spectrometry analysis because the concentration of metal ions such as copper(II), cobalt(II) and nickel(II) is generally very low. Adsorption is one of the effective preconcentration techniques. [4][5][6] In general, the adsorption technique follows the procedure to elute metal ions from the metal ions-adsorbed adsorbent.Recently, a method in which the metal ions-adsorbed adsorbent/water suspension is injected directly into a furnace, such as tungsten and graphite in electrothermal atomic absorption spectrometry, has been reported. [7][8][9][10][11][12][13] The present authors have previously discussed a directinjection method of the metal ion-adsorbed chitin/water suspension. [11][12][13] This direct-injection method would be much more useful because of the lack of necessity for the elution procedure. However, the direct-injection method needs investigations in areas such as the grain size of the adsorbent, the kind and volume of the dispersant and the dispersion time of the suspension. Furthermore, this method needs further improvements in the areas of analytical accuracy and the concentration rate.Chitosan is derived from chitin by the deacetylation of acetoamide groups with strong alkaline solutions. Chitin is one of the cellulose-like biopolymers widely distributed in nature, such as shellfish, yeast and insects. The details of chitin and chitosan chemistry were described in a previous book. 14 Especially, chitosan is well known to dissolve in some organic acids and to adsorb metal ions to form metal-chitosan complexes. There have been many reports concerning the adsorption of metal ions onto chitosan. [15][16][17][18] Moreover, Muzzarelli et al. discussed the determination of copper 19 and vanadium 20 by graphite atomic absorption spectrometry using chitosan. In these studies, an elution procedure to elute copper from copper-adsorbed chitosan 19 and an accurate operation to deposit the vanadium-adsorbed chitosan onto the graphite furnace 20 were needed.The feature of this study was to extend the chitosan's properties to the preconcentration and determination of copper(II) in water samples by tungsten metal furnace atomic absorption spectrometry. That is, the copper(II)-adsorbed chitosan was dissolved in a small volume of acetic acid, and an aliquot of the resulting solution was directly injected into the tungsten metal furnace. This proposed method is advantageous concerning the concentration factor, the analytical accuracy and the procedure compared with the suspension method, because the resulting solution is a small volume and a dispersion procedure is not need. Miniaturized coprecipitation with hydrous manganese(IV) oxide was studied for the preconcentration of sub μg dm−3 levels of molybdenum in water samples. Manganese(IV) oxide was produced by a reaction of permanganate ion with ethanol in the presence of 0.1 mol dm−3 hydrochloric acid. A nanogram amount of molybdenum in a 10 cm3 sample was quantitatively coprecipitated. The amount of manganese required was 0.1—0.40 mg for a 10 cm3 sample. The precipitate was collected on a 10 μm membrane filter, and then dissolved in 50 mm3 of 5.6 mol dm−3 nitric acid and 75 mm3 of 1% hydrogen peroxide. An 80-fold preconcentration factor was obtained by this procedure. The molybdenum concentration was determined by electrothermal atomic absorption spectrometry using a pyrolytic graphite coated cuvette. The detection limit (3σ) was found to be 8.9 ng dm−3. The proposed method was successfully applied to river water and snow samples.
