Graphene‐Based MicroRNA Transfection Blocks Preosteoclast Fusion to Increase Bone Formation and Vascularization

Abstract: The objective of this study is to design a graphene‐based miRNA transfection drug delivery system for antiresorptive therapy. An efficient nonviral gene delivery system is developed using polyethylenimine (PEI) functionalized graphene oxide (GO) complex loaded with miR‐7b overexpression plasmid. GO‐PEI complex exhibits excellent transfection efficiency within the acceptable range of cytotoxicity. The overexpression of miR‐7b after GO‐PEI‐miR‐7b transfection significantly abrogates osteoclast (OC) fusion and bo… Show more

“…The relative expression of osteoclast marker genes, including Trap , MMP‐9 , DC‐STAMP , ATP6V0D2 , c‐Fos, and NFATc1 , also demonstrated the inhibitory effects of NCF on osteoclastogenesis. DC‐STAMP participates in the fusion process, and blockage of its expression can interrupt preosteoclast fusion . Therefore, the depression of DC‐STAMP might be responsible for the inhibitory effects of NCF on cell‐cell fusion.…”

“…DC-STAMP participates in the fusion process, [23][24][25] and blockage of its expression can interrupt preosteoclast fusion. 26 Therefore, the depression of DC-STAMP might be responsible for the inhibitory effects of NCF on cell-cell fusion. In addition, NCF treatment decreased the osteoclast surface/bone surface ratio and preserved bone destruction in OVX mice.…”

“…Resorption area was quantified by the ImageJ software as previously described. 28,29 2.12 | Survival of osteoclasts After 6 days cultures of osteoclasts, RANKL was removed from the culture medium and further cultured for the indicated periods. The remaining TRAP + MNCs were counted, and the survival rate is expressed as a percentage of the remaining cells at the initial time point (0 hour).…”
