The secondary, ternary and quaternary structures of the biological ion channel Gramicidin-A are investigated by means of UV-visible and fluorescence spectroscopies, and by computational simulations in water/methanol solvent with various molar ratios. Both experiments and molecular dynamics calculations show that the two considered conformations, i.e. the head-to-head and doublestranded b-helix dimers, behave differently with regard to their stability as a function of the methanol content in the water/methanol mixture. To sum-up, calculations show that the double-stranded dimer is stabilized by methanol, even at low content, while the head-to-head dimer conformation breaks into two monomers which remain however coupled whatever the water/methanol mixture. In pure water and low methanol content, the resulting single b-helix structure is maintained due to the stabilization of the channel by water molecules and cations.

SummaryA simple, selective method is described for separation of more than 10 gramicidin components on Spherisorb ODS B, 5 I~m, 250 x 4.6 mm I. D. column, maintained at 50 ~ The mobile phase comprised methanol-water (71:29) at a flow rate of 1.0 mL min 1. Detection was by UVat 282 nm. Vahne gramicidins A, B and C were very well separated from the isoleucine gramicidins A, B and C. Four new gramicidin components were also resolved and their structures determined by liquid chromatography/mass spectrometry The names 10-methionine valine gramicidin C 4-methionine valine gramicidin A, vahne gramicidin A hydroxypropyl and isoleucine gramicidin A hydroxypropyl were proposed. Robustness of the liquid chromatography method was evaluated by performing a full factorial design experiment. The method also showed good repeatabihly, linearilyand sensitivily.
