Grafting Hyaluronic Acid onto Gold Surface to Achieve Low Protein Fouling in Surface Plasmon Resonance Biosensors

Abstract: Antifouling surfaces capable of reducing nonspecific protein adsorption from natural complex media are highly desirable in surface plasmon resonance (SPR) biosensors. A new protein-resistant surface made through the chemical grafting of easily available hyaluronic acid (HA) onto gold (Au) substrate demonstrates excellent antifouling performance against protein adsorption. AFM images showed the uniform HA layer with a thickness of ∼10.5 nm on the Au surface. The water contact angles of Au surfaces decreased fro… Show more

“…213 Furthermore, a bioorthogonal handle, such as an azide and an alkyne, was chemically introduced into polysaccharides for bioorthogonal conjugation chemistry, such as CuAAC and Staudinger reactions. 221 In order to efficiently reduce the inflammation response induced by TNF-α, HAs were conjugated to anti-TNF-α antibodies through reactions between carboxyl acids of the HAs and amines of the antibodies. 216 Although we briefly summarized the recent progress on hyaluronic acid-polymer conjugation here, it should be noted that bioorthogonal chemistries were rarely used.…”
