Gossypol, a phenolic compound produced by the pigment glands of cotton, is known to affect male reproduction. In females, exposure to gossypol has been associated with interference with the estrous cycle and embryo development. In laying hens fed cottonseed, gossypol was found to reduce egg production and weight and to cause discoloration of the egg yolk and/or albumen. It is probable that gossypol directly affects ovarian follicles, but this has not been confirmed experimentally yet. Thus, the aim of the present study was to determine if gossypol affected cultivated ovarian follicles of the chicken. Ovarian follicles of adult female chickens were cultivated with different concentrations of gossypol (0, 5, 10 and 20 μg/mL) and classified according to the stage of development as primordial, transitional, primary, secondary or antral, and as viable or atretic. The percentages of viable and atretic follicles in the ovaries cultivated for 24 hours in the presence of gossypol were significantly different from those of the controls for all types of follicles except for secondary follicles, but in all types after cultivation for 7 days. The percentage of viable follicles was higher than that of atretic follicles in ovaries cultivated without gossypol. In contrast, ovaries cultivated with gossypol showed a predominance of atretic follicles. Gossypol increased the proportion of atretic follicles at all stages of development in cultivated chicken ovaries. Thus, gossypol may affect ovarian follicular viability and maturation, which might interfere with female fertility.

Gossypol, a polyphenolic aldehyde found in cottonseed, has been shown to perturb steroidogenesis in granulosa and luteal cells of rats, pigs and cattle. However, little is known about the direct effect of gossypol on theca cell functions in any species. The present study was conducted to investigate the effect of gossypol on the steroidogenesis and the expression of genes involved in it in cultured bovine theca cells. Theca cells were isolated from healthy preovulatory follicles and were cultured in the presence of luteinizing hormone (LH) for up to 7 days. During the culture period, main steroid products of the theca cells shifted from androstenedione (A4) at day 1 to progesterone (P4) from day 2 onward. At days 1 and 7, theca cells were treated with gossypol (0-25 μg/mL) for 24 h. Gossypol inhibited LH-stimulated theca cell A4 and P4 production in a dose-dependent manner at both occasions. The viability of theca cells was not affected by gossypol at any doses used. Gossypol down-regulated expressions of steroidogenic enzymes CYP11A1, HSD3B1 and CYP17A1, but not that of LHR. These results indicate that gossypol inhibits thecal steroidogenesis through down-regulating gene expressions of steroidogenic enzymes but without affecting cell viability in cattle.

Gosipol is a substances contained in extracted cotton seed which is thought to have the antifertility ability therefore it is often used as a herbal contraceptive. The aim of this study were to assess the folliculogenesis in mice after administrated with cottonseed extract. 60 female mice strain DDY which was 14-15 weeks old and 30-35 g body weight were divided into five groups and given cottonseed extract each 0; 1,5; 2,1 and 2,7 g/kg BW for 5, 10, 15, 24, and 24 + 10 days (without cottonseed treatment). At the end of the treatment period, mice was euthanasia to observe follicular development histomorphology (each three mice of each treatment). Mice estrous status were evaluated based on the description of the vaginal smear cells with Giemsa staining. The results showed that the number of developing follicles was low (P < 0.05) compared with control after 5 days cottonseed extract administration at dose 2,7 g/kg BW that were 23 ± 3,6. At dose 1,5 and 2,1 g/kg BW the number of follicles was low after 24 days that were 25 ± 10,4 and 27 ± 3,5. Recovery effects of follicle number after cottonseed extract administration for 24 days was the best at a dose of 1,5 g/kg BW. Prolonge of estrous cycle occured in mice which were administrated the cottonseed extract of at all dose treatment. In conclusion, although the decrease in the number of developing follicles and prolonge of estrous cycles occurred after cottonseed extract administration, but these effects are reversible after the administration ended.Keywords: antifertility, cotton seed, folliculogenesis, gossypol AbstrakGosipol adalah zat yang terkandung dalam ekstrak biji kapas dan diduga mempunyai kemampuan antifertilitas sehingga sering digunakan sebagai bahan herbal kontrasepsi. Penelitian ini bertujuan mengkaji gambaran folikulogenesis pada mencit yang diberi ekstrak biji kapas. Sebanyak 60 ekor induk mencit betina strain DDY berumur 14-15 minggu dengan bobot badan 30-35 g dibagi menjadi 4 kelompok dan diberi ekstrak biji kapas masing-masing 0; 1,5; 2,1 dan 2,7 g/kg BB selama 5, 10, 15, 24 dan 24 + 10 hari (tanpa pemberian ekstrak biji kapas). Pada setiap akhir waktu pengamatan, mencit di-euthanasia untuk mengamati histomorfologi perkembangan folikel (masing-masing tiga ekor dari tiap perlakuan). Status estrus mencit dievaluasi berdasarkan gambaran sel dari ulas vagina dengan pewarnaan Giemsa. Hasil penelitian menunjukkan bahwa jumlah folikel rendah (P < 0,05) dibandingkan dengan kontrol setelah 5 hari pemberian ekstrak biji kapas dengan dosis 2,7 g/kg BB yaitu 23 ± 3,6. Pada dosis 1,5 dan 2,1 g/kg BB jumlah folikel menjadi rendah setelah 24 hari yaitu 25 ± 10,4; 27 ± 3,5. Efek pemulihan jumlah folikel setelah pemberian ekstrak biji kapas selama 24 hari paling baik pada dosis 1,5 g/kg BB. Pemanjangan siklus estrus terjadi pada mencit yang diberi ekstrak bji kapas pada semua dosis perlakuan.
