Goshuyuto, a Traditional Japanese Medicine for Migraine, Inhibits Platelet Aggregation in Guinea-Pig Whole Blood

Abstract: Abstract. The effects of goshuyuto and chotosan, traditional Japanese medicines, on collageninduced platelet aggregation were examined using guinea-pig blood. Goshuyuto at the concentration of 1,000 μg / mL inhibited collagen-induced platelet hyper-aggregation to the same degree as aspirin at the concentration of 100 μmol / L, but chotosan did not. Goshuyuto is composed of four medicinal herbs. Of them, aqueous extracts of Evodiae Fructus and Zingiberis Rhizoma inhibited platelet aggregation, but aqueous extra… Show more

“…It is prescribed, according to traditional Chinese medical practice, for the treatment of headache, abdominal pain, dysentery, amenorrhea, and postpartum hemorrhage (1). Our previous study demonstrated that goshuyuto, a traditional Japanese medicine, had an antiaggregatory effect of platelets in guinea pigs and that Evodiae Fructus was the active substance in the components of goshuyuto (2). Also, we demonstrated that goshuyuto constricted isolated rat thoracic aorta and that the constrictive effects was due to Evodiae Fructus (3).…”

“…Its two herbal components, zingiberis and evodiae, have been shown to inhibit collagen-induced platelet aggregation. Interestingly, 6-gingerol and 6-shogaol as two constituents of zingiberis can inhibit platelet aggregation by inhibition of cyclooxygenase-1 activity (Hibino et al, 2008). In another study, anti-migraine properties of Sapindustrifoliatus water extract has been linked to both inhibition of 5-HT 2B receptor and release of platelet serotonin (Arulmozhi et al, 2004).…”

“…5) In addition, we elucidated that goshuyuto inhibited the platelet aggregation of guinea pigs. 6) These results suggest that goshuyuto affects the vascular system, but there have not been any reports of pharmacological evaluation of the vascular response to goshuyuto. In the present study, we investigated the effects of goshuyuto on vascular response and its mechanism in the isolated rat aorta.…”
