Gomisin A decreases the LPS-induced expression of iNOS and COX-2 and activation of RIP2/NF-κB in mouse peritoneal macrophages

Abstract: Gomisin A (GA), a lignan component contained in the fruit of Schisandra chinensis Baillon, improves hepatic cell degeneration, vasodilatory activity and insulin sensitivity. These effects also impact the immune system, including various inflammatory mediators and cytokines. In this study, the anti-inflammatory effect of GA on lipopolysaccharide-stimulated mouse peritoneal macrophages was studied. Pretreatment with GA attenuated the expression of receptor-interacting protein 2 (RIP2) and IκB kinase-β (IKK-β) as… Show more

“…10,11 Gomisin A is a small-molecular-weight component contained in the fruit of Schisandra chinensis Baillon, Which has anti-inflammatory and antioxidant activities. [12][13][14] Previous studies suggest that the antioxidant compounds from Schisandra chinensis including schisandrin B, schisandrin C, gomisin J, and gomisin A attenuate oxidative stressinduced by excessive ROS. 15,16 However, the effects of gomisin A on age-related anti-inflammation and anti-oxidation are still unknown.…”

“…When macrophages are stimulated with LPS, the production of pro-inflammatory cytokines is markedly increased (Jeong et al, 2014). To study the effect of SKL on the production of proinflammatory cytokines, we performed ELISA.…”

“…The high amount of NO produced following exposure to LPS plays a vital role in increasing inflammatory reactions (Jeong et al, 2014). To examine the effect of SKL on LPS-induced NO production, we carried out Griess reagent method.…”

“…The NF-κB agonist LPS (10 μg/mL) and inhibitor PDTC (10 μM) were used to activate or suppress NF-κB in ATO-treated cells. Previous studies showed that LPS and PDTC at these two dosages can sufficiently activate or inhibit NF-κB without affecting cell growth in many cell lines, including HepG2 cells (Jeong et al, 2014;Song et al, 2014). The result demonstrated that ATO dosedependently decreased cell viability (Fig.…”
