Gold nanoparticles delivered miR-375 for treatment of hepatocellular carcinoma

Abstract: MiR-375 is a tumor suppressor miRNA that is downregulated in hepatocellular carcinoma (HCC). However, due to the lack of effective delivery strategies, miR-375 replacement as a therapy for HCC has not been investigated. In the present study, we have developed a straightforward strategy to deliver miR-375 into HCC cells by assembling miR-375 mimics on the surface of AuNPs and forming AuNP-miR-375 nanoparticles. AuNP-miR-375 exhibits high cellular uptake and preserves miR-375′s activities to suppress cellular pr… Show more

“…Intratumoral injection of miR-375 nanoparticle formulation markedly reduced tumor growth of HepG2 xenograft masses. These findings were confirmed in a second HCC mouse model obtained by hydrodynamic injection of AKT/Ras oncogenes and tail-vein administration of AuNP-miR-375 showing a consistent increase of apoptotic markers and a reduction of tumor nodules and hepatocyte proliferation [24].…”

“…MicroRNAs are a class of highly conserved endogenous ncRNAs that are characterized by the single-stranded structure and low molecular weight (19)(20)(21)(22)(23)(24)(25) nucleotides in length). 12,13 Numerous miRNAs have been identified as the important modulator for various cellular events, including cell differentiation, proliferation, apoptosis and oncogenesis, through negatively regulating the expression of target genes at posttranscriptional level.…”

“…These particles can be used as contrast agents for diagnosis by MRI. Gold NPs main accumulation site is in the liver, and thus, it is a promising biomaterials for the treatment of liver cancer [74,75]. Gold NPs are usually capped with either sodium citrate, polyamidoamine dendrimers (PAMAM), or L-aspartate to stabilize the particles and their cellular uptake is usually mediated by clathrin dependent endocytosis.…”
