Gold nanoparticle-based paper sensor for multiple detection of 12 Listeria spp. by P60-mediated monoclonal antibody

Abstract: The genus of Listeria consists of heterologous species and its presence in the food chain is an indicator of poor hygiene. However, a portable and simple paper sensor for detection of listeria spp. with high accuracy was still unknown. In this study, we prepared a pair of monoclonal antibodies (mAbs) that specifically recognize the P60 protein on the cell surface of Listeria spp. The selected pair was found to be sensitive to both the P60 protein and cell body of the genus Listeria. On this basis, a rapid pape… Show more

“…The incorporation of gold nanoparticles in LFSA is a common practice in more recent approaches (29,67,70,109). Gold nanoparticles (AuNPs) were widely used for antibody conjugation ascribed to their high surface-to-volume ratios, increasing the amount of CFU/mL of E. coli O157:H7 as opposed to 10 4 CFU/mL in popcornlike probes and 10 5 CFU/mL in large-sized flowerlike probes.…”

“…In 2017, Wang et al () established GNP‐based LFAs (~15 nm GNPs) for multiple detection of 12 Listeria spp . With their technique, they could detect broadly cross‐reacted Listeria at a sensitivity of ~10 3 –10 4 CFU mL −1 .…”

“…The selected hybridoma cells were expanded and injected into BALB/c mice to produce mAbs (Deng et al, 2012;Suryoprabowo, Liu, Peng, Kuang, & Xu, 2014). The ascites were harvested and purified using the caprylic acid-ammonium sulfate precipitation method (Kong et al, 2017;Wang et al, 2017). The purified antibody solution was stored at −20 o C until further use.…”
