GntR-Type Transcriptional Regulator PckR Negatively Regulates the Expression of Phosphoenolpyruvate Carboxykinase in Corynebacterium glutamicum

Abstract: The pck (cg3169) gene of Corynebacterium glutamicum encodes a phosphoenolpyruvate carboxykinase (PEPCK). Here, a candidate transcriptional regulator that binds to the promoter region of pck was detected using a DNA affinity purification approach. An isolated protein was identified to be PckR (Cg0196), a GntR family transcriptional regulator which consists of 253 amino acids with a mass of 27 kDa as measured by peptide mass fingerprinting. The results of electrophoretic mobility shift assays verified that PckR … Show more

“…The binding site of IolR within the pck promoter is located far upstream of the TSS, and the about-3-fold-lower mRNA level and the about-2-fold-lower specific PEP carboxykinase activity in the ⌬iolR mutant than in the WT indicate that IolR acts as an activator. These results are entirely different from those of a recent study by Hyeon et al (77). Those authors described Cg0196 (IolR) as a novel regulator which negatively regulated pck expression when the cells were grown on glucose as the carbon source, and thus they designated the Cg0196 protein PckR.…”

“…Of these proteins, SPD_0064 was of particular interest because it belongs to the GntR family transcription regulator ( Fig. S1 ) and may have the capacity to bind glucose 27 . Examination of the localization of SPD_0064 gene revealed that it is located approximately 25,000 nt apart from the cps locus in S. pneumoniae D39.…”

“…The GntR family is one of the most abundant and widely distributed groups of transcriptional regulators in bacteria [ 34 ]. It was reported that the gntR gene acts to repress transcription of itself as well as a series of genes immediately adjacent to it [ 35 , 36 ]. Mutation of gntR gene resulted in increased LAAO activity, indicating a downregulation role of gntR gene in Pseudoalteromonas sp.…”
