Glypicans define unique roles for the Hedgehog co-receptors boi and ihog in cytoneme-mediated gradient formation

Abstract: The conserved family of Hedgehog (Hh) signaling proteins plays a key role in cell-cell communication during development, tissue repair and cancer progression, inducing distinct concentration-dependent responses in target cells located at short and long distances. One simple mechanism for long distance dispersal of the lipid modified Hh is the direct contact between cell membranes through filopodia-like structures known as cytonemes. Here we have analyzed in Drosophila the interaction between the glypicans Dall… Show more

“…Moreover, all signals, including those that are not known to be lipidated (e.g., BMPs and many FGFs), can interact with membrane-anchored proteoglycans, which can restrict free signal dispersion and induce biphasic signaling activation and inhibition [61][62][63] . Glypicans also can control cytoneme stability [64][65][66] . Therefore, our findings showing how GPI anchoring enables Bnl to direct source and recipient cells to reciprocally coordinate with each other by cytonemes provide important new insights into how other signal retention strategies might control signaling and morphogenesis.…”

“…The HSPGs Dally and Dlp cluster on the cell surface of cultured Drosophila cells and Dlp clusters on Hh-transporting cytonemes [142]. In the Drosophila wing disc, Dally and Dlp interact with and stabilize ectopically expressed Ihog on cytoneme membranes by slowing cytoneme elongation and retraction velocities [143]. Similarly, BOC and CDON enrich in microdomains along portions of chick limb bud cytonemes that remain static, suggesting that they bind to extracellular components to stabilize the structures as they extend [62].…”
