Glycosylation of hemoglobin in vitro: affinity labeling of hemoglobin by glucose-6-phosphate.

Abstract: To determine the mechanism for the formation of hemoglobin Alc (Hb Ale) in vivo, we incubated human hemoglobin with glucose and metabolites of glucose. ['4CJGlu-cose-6-phosphate (G6P) reacted readily with deoxyhemoglobin, and formed a covalent linkage. The reaction rate was considerably reduced in the presence of carbon monoxide or 2,3-diphosphoglycerate (2,. Purified G6P hemoglobin had a lowered oxygen affinity and decreased reactivity with 2,3-DPG compared to Hb A. G6P behaved as a 2,3-DPG analog and reacted… Show more

“…On the basis of precursor studies in erythrocyte hemolysates, it was suggested that the synthesis of HbAIc proceeds via the interaction of HbA with glucose-6-phosphate, yielding a HbA-glucose-6-phosphate intermediate, which in turn is dephosphorylated to HbAl, (6,7). HbAIb was proposed as the intermediate by Stevens et al (6), whereas Haney and Bunn (7) suggested either HbAia or HbAIb. However, in a recent ment (9)(10)(11).…”

“…Clinical interest in the heterogeneity of hemoglobin A has been stimulated by the observation that posttranslational adducts of the protein form spontaneously with reducing sugars (1,2). The resulting minor hemoglobins (HbAia-c)l have been used increasingly This work was presented in part at the 5th Biennial International Symposium on Alcoholism, 9-13 June 1980, Cardiff, Wales.…”

“…However, addition of a phosphate at Val-1 (β) does not seem to be enough to exhibit such impact on the oxygen affinity of Hb. Affinity labeling of Val-1(β) with glucose-6-phosphate does not reduce the oxygen affinity of Hb (54) to the extent that is seen with G3P or pyridoxal phosphate. The structural features of these added groups seem to make major contribution towards this effect.…”
