Glycoproteins Associated with Hepatitis B Antigen

Abstract: The total carbohydrate content of purified hepatitis B antigen (HB Ag) ranged between 3.6 and 6.5% as determined by the phenol-sulfuric acid method. Disc-sodium dodecyl sulfate PAGE of purified HB Ag followed by periodate-Schiff staining revealed one major and two minor glycoproteins with molecular weights of 22,000,27,000 and 32,000 daltons, respectively. Both ad and ay subtypes of HB Ag contained these glycoproteins.

“…Although Dreesman et al (18,22) reported finding small amounts of two peptid es (69,000 and 105,000 daltons) in HBsAg/ ayw that were not present in HBsAg/ adw, no difference has been fo und in polypeptides of different subtypes by investigators (Table I). Shih & Gerin (88) fo und the sam e sizes, num bers, and relative amounts of polypeptides in purified preparations of HBsAg with SUbtypes adw, ayw, adr (Table 1).…”

“…Early studies of purified 22-nm particles by using sodium dodecyl sulfate (SDS)-polyacrylamide gel electrophoresis to separate peptides identified as few as two (96) or three (28) major polypeptides with sizes between 26,000 and 40,000 daltons (see Table 1). More recently, as many as seven (25,87) or nine (18,22) peptides ha ve been identified by Coomasie blue stain ing of SDS-polyacrylamide gels with size ranges from 23,000-97,000 daltons and from 19, 000-120,000 daltons, respectively, in two representative studies (Table 1). Two (26,88) or three (18) polypeptides have been shown to stain with periodic acid-Schiff, suggesting they are glycoproteins.…”

“…The molecular masses of the polypeptides identified in HBAg, which has a particle weight of approximately 7 X 10-18 g (8), add up to 320,000-490,000 daltons (9,10), corresponding to 2550 to 3935 amino acids as calculated from the amino-acid composition (8). The lower and upper numbers correspond to the d and y subtypes of HBAg, respectively.…”

“…When mixtures of 0.5 ml of marguerite extract and 0.5 ml of HBs antigen sensitized erythrocytes or 0.5 ml of pure HBs antigen (100 r/ml) were incubated at 5 OC overnight, the agglutinin activity disappeared. However, when the extract was [4] reported that the HBs antigen contains 3 . 6 4 5 % of glycoprotein, and Neuruth et al [5] also reported that N-acetylneuraminic acid is contained in H B s antigenic substances.…”
