Glycoprotein H and α4β1 Integrins Determine the Entry Pathway of Alphaherpesviruses

Abstract: c Herpesviruses enter cells either by direct fusion at the plasma membrane or from within endosomes, depending on the cell type and receptor(s). We investigated two closely related herpesviruses of horses, equine herpesvirus type 1 (EHV-1) and EHV-4, for which the cellular and viral determinants routing virus entry are unknown. We show that EHV-1 enters equine epithelial cells via direct fusion at the plasma membrane, while EHV-4 does so via an endocytic pathway, which is dependent on dynamin II, cholesterol, … Show more

“…Moreover, tyrosine kinase activity is necessary for the aggregation and fusion of caveolae or caveolar vesicles (54). Therefore, it is not surprising that a number of viruses gain access to host cells by nonclassical endocytic pathways that are all dependent on the action of dynamin and tyrosine kinases but differ in other factors involved (55)(56)(57)(58). Here, we studied the role of caveola-dependent endocytosis in virus-induced MHC-I downregulation.…”

“…Those were BACs of EHV-1 strain Ab4 (33) and EHV-4 strain TH20p (34), as well as modified BACs EHV-1_gB4, EHV-4_gB1, revertant EHV-1_gB1r (20), EHV1_gD4, EHV-4_gD1 (35), EHV-1⌬US3, and EHV-1 that contained US3 of EHV-4 (EHV-1_US3_4) in lieu of authentic US3, fully rescuing the parental EHV-1 phenotype and functioning as revertant for this study (A. Proft and W. Azab, unpublished data). monomeric red fluorescent protein (mRFP1)-labeled EHV-1 was previously constructed by inserting mRFP1 into VP26 of EHV-1 strain RacL11 (36). Viruses were grown on equine dermal (ED) cells.…”

“…While the specific role of lipid rafts in alphaherpesvirus entry is not known, pseudorabies virus (PRV) localizes in juxtaposition with GM1, a lipid raft marker (49), and herpes simplex virus 1 (HSV-1) gB associates with lipid rafts during virus entry (50). Additionally, cholesterol is important for entry of alphaherpesviruses (49)(50)(51)(52)(53). BoHV-1 requires cell membrane cholesterol for entry into Madin-Darby bovine kidney (MDBK) cells and viral envelope cholesterol for infectivity (54).…”
