Glycollate oxidation by thylakoids of the cyanobacteria Anabaena cylindrica, Nostoc muscorum and Chlorogloea fritschii

Abstract: The intracellular distribution of glycollate dehydrogenase EC 1.2.1.17 has been investigated in extracts of the cyanobacteria (blue-green algae) Anabaena cylindrica, Nostoc muscorum and Chlorogloea fritschii. Most of the enzyme activity was associated with a chlorophyll-containing cell-free pellet, which also exhibited Photosystem I and II activities. Sucrose density gradient centrifugation of this washed pellet resulted in the formation of a green band within which maximal chlorophyll concentration and enzymi… Show more

“…The procedure for the production of the chlorophyll-containing pellets was essentially that for thylakoid isolation from the cyanobacteria, Oscillatoria chalybea [16] Anabaena cylindrica, Nostoc muscorum and Chlorogloea fritschii [17].…”

“…Individual pigments were resolved by thin layer chromatography and the spots were identified by their characteristic colours with staining reagents and by their RF values (Stahl, 1967). The activities of photosystems I1 and I, and NADP-photoreduction, were measured as described by Codd & Sallal (1978).…”

“…The enzyme activities retained in washed pellet produced after 35,000 x g for 30 min were 60% to 70% of the original, while 6% to 7% of the original activities were found in the supernatant (Table 1). The procedure for the isolation of the Glutamine synthetase in Chlorogloeopsis chlorophyll-containing pellet was essentially that described for thylakoid isolation (Codd & Sallal 1978). Figure 1 shows the distribution of pglutamyl transferase activity of glutamine synthetase in fractions following sucrose density centrifugation of both thylakoid membranes isolated by high speed centrifugation and the crude cell extract.…”
