Glycolipid-peptide vaccination induces liver-resident memory CD8+ T cells that protect against malaria

Abstract: Liver resident-memory CD8+ T cells (TRM cells) can kill liver-stage Plasmodium-infected cells and prevent malaria, but simple vaccines for generating this important immune population are lacking. Here, we report the development of a fully synthetic self-adjuvanting glycolipid-peptide conjugate vaccine designed to efficiently induce liver TRM cells. Upon cleavage in vivo, the glycolipid-peptide conjugate vaccine releases an MHC I-restricted peptide epitope (to stimulate Plasmodium-specific CD8+ T cells) and an … Show more

“…Resident memory T cells are crucial for local immunity and recall response, which is considered more potential subsets in vaccine designation (92). A recent study shows that induction of iNKT cell activation can promote the generation of liver-resident memory CD8 + T cells, which prevents malaria (93). In naïve mice raised in specific pathogen-free and germ-free conditions, a small CD8 + T-cell population displaying memorial phenotypes is named "virtual memory" CD8 + T cells (CD8 + T VM ).…”

“…Oxime ligation between carbonyl-containing compounds (i.e., aldehydes or ketones) and peptides functionalized with an aminooxy group is a proven way to efficiently generate glycolipid-peptide conjugate vaccines. 20,21,49 Conditions promoting oxime bond formation include the use of aniline catalysts under mildly acidic conditions (∼pH 3−5). 50 To investigate this ligation approach, the corresponding G0 vaccine construct was synthesized whereby α-GalCer amine 5 was reacted with the activated ester of 8-oxo-nonanoic acid (23) to give α-GalCer ketone 24 (Scheme 5).…”

“…RAS vaccination is considered the gold-standard for the generation of liver Trm and protection from malaria. This finding led to development of liver Trm cell generating vaccines that provide sterile protection from malaria [379][380][381][382] . Most notable are, "prime and trap" vaccination strategies that use either DNA vaccination or peptide-linked anti-CLEC9a…”

“…Trm cells in the brain produce a barrier that protects the brain from LCMV infection [406] . NKT cells can be exploited to generate powerful liver Trm responses that can protect from malaria [379,380] . Interestingly, NKT have been considered Trm cells in many contexts, as they are typically non-circulating [352] .…”
