Glycogen depletion in intrafusal fibres in rats during short‐duration high‐intensity treadmill running

Abstract: The results suggest that during brief-intensity exercise, as the glycogen content of type IIA fibres is reduced earlier than that of type I fibres, bag2 fibres are most important early in this type of exercise.

“…Midventricular sections were stained with Schiff Reagent (Polyscientific, Bay Shore, NY) to identify glycogen in hearts [periodic acid-Schiff (PAS) staining] (30). The specificity of glycogen staining was confirmed by treating sections with diastase to digest tissue glycogen, followed by regular PAS staining (30). Sectioned heart tissues were stained for DNA fragmentation by a terminal deoxynucleotidyl transferase-mediated dUTP-biotin nick end labeling (TUNEL) staining protocol according to the manufacturerʼs instructions (R and D Systems, Minneapolis, MN).…”

“…Serial sections in the mid-belly of the muscles were prepared for histochemical procedures, myosin ATPase [16], succi-nate dehydrogenase (SDH, [17]), alpha-amylase-periodic acid schiff (PAS, [14]) and IMCL (OR; oil red O staining, [18] [19]. The evaluation of the histochemical profile of each fiber was estimated by optical density based on a computer system [14]. In brief, two criteria, complete transmission without a slide glass (130 ± 0.5) and with one (120 ± 0.5), were used to adjust to constant light absorbance (0 = complete absorbance, 250 = complete transmission).…”

“…IMCL of soleus muscle is reportedly higher than that of the other skeletal muscles [2] [13], and IMCL of type I is higher than type II fiber types [8], while soleus muscle is mainly composed of type I fibers [14]. The objective of this study was to examine the effect of exercise training on glycogen and IMCL content of soleus muscle in NASH model rats.…”
