Glycerol‐3‐phosphate dehydrogenase is induced by glucocorticoids in hepatocytes and hepatoma cells in vitro

Abstract: Previous studies have shown that cytosolic glycerol-3-phosphate dehydrogenase (GPDH; EC 1.1.1.8) can be induced by glucocorticoids in mammalian brain, mammary gland, and thymus, but it was thought that no induction occurred in liver. We report here that GPDH is induced by glucocorticoids in several lines of hepatoma cells and in rat hepatocytes cultured in vitro. When rat hepatoma cells of clone FU5AH were exposed to 3 microM hydrocortisone (HC) for 3 days, GPDH specific activity increased greater than sixfold… Show more

“…Aliquots of the unfractionated brain homogenates from the first step in the myelin isolation procedure were stored at -70°C for subsequent determinations. A portion of the aliquot was centrifuged at 30,900 g for 1 h, and the supernatant was assayed for GPDH activity, as described by Meyer et al (1983). CNP activity was assayed in unfractionated homogenates, following the procedure of Prohaska et al (1973).…”

“…Cytosolic GPDH activity and gene expression are increased during the active phase of glycerol accumulation in rainbow smelt following the natural thermal cycle (12,13), GPDH activity increases in liver of fish subjected to acute decreases in water temperature (6), and GPDH mRNA decreases in fish subjected to acute temperature increase (8). GPDH activity is stimulated in rat hepatoma cell lines and transformed rat hepatocytes after 3 days of in vitro culture with glucocorticoids (14), and GPDH activity and mRNA level are stimulated by glucocorticoids in the rat brain (2). Also, in mouse liver, the GPDH gene has a functional peroxisome proliferator-activated receptor (PPAR) response element in its promoter that is a direct target of PPAR␣ (21).…”

“…Interestingly, it has been shown that this enzyme can be induced by glucocorticoids in several mammalian tissues (Meyer et al, 1983). The spot intensity in corticosterone-treated samples was also increased (1.7-fold) compared to controls.…”
