Glutathione S-transferase class ? in French alcoholic cirrhotic patients

Abstract: The lack of glutathione S-transferase mu (GST mu) was examined in 45 healthy French Caucasians and 45 alcoholic cirrhotic French Caucasians: microsamples of blood were taken and DNA amplified by the polymerase chain reaction. We have concluded that there is no relationship between this genotype and the development of alcoholic cirrhosis in these heavy consumers of ethanol.

“…Detailed information of the PCR assays can be found elsewhere [Groppi et al, 1991]. Genotypes of GSTM1 were determined by the presence or absence of PCR product, according to the method of Groppi et al [1991]. Two primers, 5Ј-GAAGGTGGCCTC-CTCCTTGG-3Ј and 5Ј-AATTCTGGATTGTAGCAGAT-3Ј, were used for PCR.…”

“…Subjects were divided into three groups corresponding to homozygous wild (Ile/ Ile), heterozygous (Ile/Val) and homozygous mutant (Val/Val) types. The GSTM1 genetic polymorphism was determined by the method of Groppi et al (1991). The presence or absence of an ampli®cation product results in two subgroups corresponding to GSTM1 present or GSTM1 null.…”

“…GSTM1-A and GSTM1-B, and by one null allele, viz. GSTM1 0 (Groppi et al 1991;Harada et al 1992). The latter does not produce a protein product because of an expanded deletion (Seidegard et al 1988).…”
