Introduction: The goal of this study was to assess the influence of vaccination against enteric redmouth disease on oxidative stress biomarkers and antioxidant defence in the muscle tissue of rainbow trout (Oncorhynchus mykiss Walbaum) vaccinated against Yersinia ruckeri in the first and second month after immunisation. Material and Methods: Healthy fish were vaccinated orally with inactivated whole cells of a virulent strain of Y. ruckeri. One and two months after immunisation the muscle samples were collected. Results: No significant difference was noted in lipid peroxidation level in either the first or second month after vaccination, while aldehydic and ketonic derivatives of oxidatively modified proteins (OMB) in the vaccinated group were significantly lower in the second month compared to those in the first month after vaccination (P < 0.05). The content of ketonic derivatives of OMB in muscles in the first month after immunisation was higher compared to untreated control. All these culminated in a depletion of glutathione peroxidase (GPx) activity and low level of total antioxidant capacity (TAC). Conclusion: Correlations between catalase activity and lipid peroxidation and TAC confirmed the pivotal role of catalase in antioxidant defence during immunisation. From a broader perspective, it is suggested that immunisation of fish with Yersinia vaccine is associated with induced free radical formation and oxidative stress. Free radicals would therefore be at least partially responsible for the induction of both humoral and cellular elements of the immunity and increased protective immunity against Y. ruckeri infection.

Ascorbate pro-drug theory promising possible effective and nontoxic treatment is intensively examined and centered on the importance of both extracellular and intra-cellular effects of ascorbate. In this study, we sought to determine the in vitro cell responsiveness in a plasma free milieu with or without an extracellular catalyst and/or hydrogen peroxide. This project brought to light how ascorbic acid would react to in a Red Blood Cell (RBC) suspension by using the two anti-oxidant pathways. We demonstrated that while ascorbate doesn't require an extracellular metal catalyst to induce cell damage via the lipid peroxidation pathway, the protein-centered metal pathway could be enhanced by a divalent cation, hydrogen peroxide or both. The current lack of understanding of ascorbate potential mechanism excites us to propose some insights for the vitamine C pharmacologic dose pro-oxidant activity.

Antioxidant defense system and oxidative stress biomarkers were determined in the liver tissue from males and females of sea trout (Salmo trutta m. trutta L.) affected by Aeromonas hydrofila infection causing ulcerative dermal necrosis (UDN) syndrome. Histological results from hepatic parenchyma in UDN-affected trout show that cytoplasmic vacuolation of hepatocytes, loss of normal basophilia by cytoplasm, presence of pigment deposition in the parenchyma are a signs of liver deterioration. Impaired circulation in the liver of UDN-affected females manifests in oedema and an occurrence of a large number of erythrocytes in the sinusoids and the intercellular space. In both UDN-affected males and females, protein oxidation showed higher values as compared to the healthy trout. UDN syndrome caused a decrease of antioxidant enzyme activities. This might be due to inactivation of the above-mentioned enzymes by the end products of protein oxidation. The importance of the glutathione-mediated antioxidant defense system in UDN-induced oxidative stress was demonstrated. Results showed that UDN syndrome led to an oxidative stress in liver tissue able to increase of the oxidatively modified protein level.
