Glutathione Peroxidase GPX1 and Its Dichotomous Roles in Cancer

Abstract: As the first identified selenoprotein, glutathione peroxidase 1 (GPX1) is a widely and abundantly expressed antioxidant enzyme. GPX1 utilizes glutathione as a substrate to catalyze hydrogen peroxide, lipid peroxide, and peroxynitrite, thereby reducing intracellular oxidative stress. The GPX1 gene is regulated at transcriptional, post-transcriptional, and translational levels. Numerous case-control studies and meta-analyses have assessed the association between a functional genetic polymorphism of the GPX1 gene… Show more

“…Antioxidant GPX1 was reported that it played a vital role in normal cell growth and adaptive pathological reaction, such as apoptosis or inflammation 67 . However, the aberrantly increased expression levels of GPX1 in many cancers were closely related to tumorigenesis and progression 55 . Previous researches had already revealed that the abnormally increased GPX1 activity could decrease tumor sensitivity toward ROS‐generating anticancer drugs 68,69 .…”

“…Among them, the expression levels of six genes (PECR, OLAH, ACOT11, ACAT2, NUDT19, PTGIS) were significantly upregulated and 4 genes (GPX1, ADH5, PTGR1, ACADS) were downregulated in UCEC samples compared to normal tissues, which have been validated by PCR experiment. Substantial evidence revealed that most of these FAMGs functioned as oncogenes or tumorsuppressor genes in various cancers (29)(30)(31)(32)(33)(34)(35)(36). We also determined the prevalence of genetic mutation in UCEC samples, as well as CNV alterations.…”

“…As GPXs detoxify ROS, it is therefore unsurprising that they are involved in detoxifying ROS generated by cancer cells. GPX1, a Se-sensitive selenoprotein whose complex roles in cancer have already been reviewed in detail [ 91 ], was reported to be involved in protective autophagy in pancreatic ductal adenocarcinoma (PDA) cells during glucose deprivation. GPX1 overexpression sensitized PDA cells to caspase-dependent apoptosis [ 86 ].…”
