Glutathione Homeostasis and Functions: Potential Targets for Medical Interventions

Abstract: Glutathione (GSH) is a tripeptide, which has many biological roles including protection against reactive oxygen and nitrogen species. The primary goal of this paper is to characterize the principal mechanisms of the protective role of GSH against reactive species and electrophiles. The ancillary goals are to provide up-to-date knowledge of GSH biosynthesis, hydrolysis, and utilization; intracellular compartmentalization and interorgan transfer; elimination of endogenously produced toxicants; involvemen… Show more

“…It cycles between two forms: the reduced thiol form, GSH, and the oxidized form, GSSG and is converted back to reduced form. [58][59][60] Doxorubicin treatment reduced the GSH concentration in a dose and time dependent manner, whereas treatment of mice with naringin before or after DOX administration elevated it significantly. The naringin pretreatment afforded better protection against DOX induced depletion than the post-treatment.…”

“…The reasons remain still unclear, but might be partially explained by the synthesis of glutathion (GSH) in the intestine. The homeostasis of GSH in animal tissues is an important guarantee for its function (Lushchak 2012). In addition, both γ-glutamylcysteine synthetase (γ-GCS) and γ-GT participated in GSH synthesis in endothelial cells (Moellering et al 1999).…”

“…GSH is consumed in various ways, such as by oxidation, conjugation and hydrolysis. GSH can be directly oxidized by RONS, act as a substrate for GSH‐dependent enzymatic reactions and conjugate with endogenous and exogenous electrophiles 172. GSH is distributed to intracellular organelles including the ER, nucleus and mitochondria 173.…”
