Glucosylceramide Biosynthesis is Involved in Golgi Morphology and Protein Secretion in Plant Cells

Abstract: Lipids have an established role as structural components of membranes or as signalling molecules, but their role as molecular actors in protein secretion is less clear. The complex sphingolipid glucosylceramide (GlcCer) is enriched in the plasma membrane and lipid microdomains of plant cells, but compared to animal and yeast cells, little is known about the role of GlcCer in plant physiology. We have investigated the influence of GlcCer biosynthesis by glucosylceramide synthase (GCS) on the efficiency of prote… Show more

“…In MDR1-MDCK cells, cell surface MDR1 colocalized with globotriaosyl ceramide (Gb 3 ), and a soluble analog of Gb 3 proved an MDR1inhibitor (De Rosa et al 2008). In plant cells, secretory products accumulate in swollen Golgi after GSL synthesis inhibition, and overexpression of such products results in a compensatory increase in GlcCer and sterol synthesis (Melser et al 2010). This is consistent with evidence that protein cargo sorting during Golgi transit is based on lipid gradients, rather than time-dependent cysternal transit (Patterson et al 2008).…”

“…Sphingolipids are essential components of the plasma membrane and nanoscale subcompartments of the plasma membrane, known as membrane rafts [1,2]. Some of these lipids are also found in the endomembrane system where they are involved in maintaining homeostasis through regulating cell secretion activity [3][4][5]. Beside their structural role, sphingolipids were further shown to participate in the control of autophagy and cell fate under stress conditions in animal and yeast cells [6,7].…”
