Glucosylation Activity and Complex Formation of Two Classes of Reversibly Glycosylated Polypeptides

Abstract: Reversibly glycosylated polypeptides (RGPs) have been implicated in polysaccharide biosynthesis. In plants, these proteins may function, for example, in cell wall synthesis and/or in synthesis of starch. We have isolated wheat (Triticum aestivum) and rice (Oryza sativa) Rgp cDNA clones to study the function of RGPs. Sequence comparisons showed the existence of two classes of RGP proteins, designated RGP1 and RGP2. Glucosylation activity of RGP1 and RGP2 from wheat and rice was studied. After separate expressio… Show more

“…Given their Golgi localization and glycosylation activity, RGPs have been implicated in polysaccharide biosynthesis, particularly cell wall and starch synthesis. 15,23 However there is little direct evidence to support these hypotheses. The strongest evidence for a role in cell wall synthesis derives from studies of pollen development in Arabidopsis.…”

“…Individual expression of OsUAM1 and UAM2 in E. coli results in homo-hexamers. 25) These suggests that UAM forms homo-and hetero-complexes. In the presents study, the molecular weight of CrUAM was determined to be 39 kDa by SDS-PAGE and about 700 kDa by size-exclusion chromatography (Figs.…”
