Glucose uptake by the cellulolytic ruminal anaerobe Bacteroides succinogenes

Abstract: Glucose uptake by Bacteroides suceinogenes S85 was measured under conditions that maintained anaerobiosis and osmotic stability. Uptake was inhibited by compounds which interfere with electron transport systems, maintenance of proton or metal ion gradients, or ATP synthesis. The most potent inhibitors were proton and metal ionophores. Oxygen strongly inhibited glucose uptake. Na+ and Li+, but not K+, stimulated glucose uptake. A variety of sugars, including atmethylglucoside, did not inhibit glucose uptake. On… Show more

“…An ABC protein associated with a xylan metabolism operon has been identified based on DNA sequence data in R. flavefaciens (21). By contrast, F. succinogenes utilizes a Na ϩ electrochemical gradient for uptake of glucose (195) and cellobiose (412), and the Na ϩ requirement for growth on cellulose suggests that Na ϩ may be the driving force for uptake of cellodextrins as well (740).…”

“…In addition, Russell also showed that valinomycin collapsed ⌬⌿ in F. succinogenes (22). Furthermore, when used alone CCCP and monensin inhibited respectively glucose and cellobiose uptake by F. succinogenes (18,19).…”

“…More recently, Simor et al (16) compared the effect of Dy(TTHA) 3Ϫ , Tm(DOTP) 5Ϫ , and Dy(PPP i ) 2 7Ϫ on isolated perfused rat hearts and showed that Tm(DOTP) 5Ϫ was the most appropriate under their experimental conditions. F. succinogenes is a major rumen bacterium which requires sodium for growth and for glucose and cellobiose uptake (17)(18)(19), suggesting that sodium may play an important role in the bioenergetics of the bacterium. In order to measure sodium transmembrane gradient in this bacterium and its variation according to extracellular sodium concentrations and pH, we developed a methodology to measure accurately Na ϩ internal concentrations in F. succinogenes S85 by 23 Na NMR.…”
